Microbial culturomics to isolate halophilic bacteria from table salt: genome sequence and description of the moderately halophilic bacterium Bacillus salis sp. nov.

Abstract: Bacillus salis strain ES3T (= CSUR P1478 = DSM 100598) is the type strain of B. salis sp. nov. It is an aerobic, Gram-positive, moderately halophilic, motile and spore-forming bacterium. It was isolated from commercial table salt as part of a broad culturomics study aiming to maximize the culture conditions for the in-depth exploration of halophilic bacteria in salty food. Here we describe the phenotypic characteristics of this isolate, its complete genome sequence and annotation, together with a comparison wi… Show more

“…In 2016, three stool samples were collected with the aim of studying halophilic bacteria involved in human gut functioning [ 12 ]. Marseille-P3515 T and Marseille-P3516 T were isolated from the stool samples of a 17-year-old boy and a 10-year-old girl, respectively, both living in Ndiop, a rural area in Senegal.…”

“…In the future, halogenated drugs may have increased action against drug-resistant bacteria [136,137]. OMIC technologies present new potential for the discovery of exclusive properties and/or novel biomolecules derived from halophiles in the future [138,139] as a result of recent findings of halophilic bacteria, even from terrestrial environments [140].…”

OMIC technologies present new potential for the discovery of exclusive properties and/or novel biomolecules derived from halophiles in the future [138,139]as a result of recent findings of halophilic bacteria, even from terrestrial environments [140]. Further research is needed to report on how halophilic microorganisms evolved during the early phases of evolution of life on earth, as well as how they diversified and spread around the world.…”

“…In culturomics research, various culture conditions are applied: various broth and solid media, blood culture bottles, selective agar media, different temperatures and times of incubation from a few days to months, various pH, oxygen and pressure sensitivity. To date, culturomics analysis has been used in taxonomic identification of clinical microbes and in the discovery of new species (Khelaifia et al, 2016;Masucci et al, 2017;Bilen et al, 2018;Seck et al, 2018). reported the description of a new bacterial species using culturomics.…”

“…In the study of Masucci et al (2017), culturomics was used for isolation of Clostridium difficile from faecal samples obtained from healthy donors of faecal microbiota transplantation (FMT). Further, Seck et al (2018) used a culturomics study to isolate the halophilic bacterium Bacillus saliss sp. nov. from table salt using high-salt conditions to cultivate halophilic bacteria from environmental samples.…”
