Microbeam evolution: from single cell irradiation to pre-clinical studies

Abstract: The development of radiation microbeams has been a valuable tool for the exploration of fundamental radiobiological response mechanisms. The strength of micro-irradiation techniques lies in their ability to deliver precise doses of radiation to selected individual cells in vitro or even to target subcellular organelles. These abilities have led to the development of a range of microbeam facilities around the world allowing the delivery of precisely defined beams of charged particles, X-rays, or electrons. In a… Show more

“…The treatment of radio-resistant cancers using Microbeam Radiation Therapy (MRT) is based on the possibility of improved therapeutic ratios resulting from reduced normal tissue complications while maintaining high tumour control probability when exposed to high, spatially fractionated doses. Whilst MRT is currently in the pre-clinical stage, there are many studies demonstrating the efficiency of the MRT treatment modality 1018 . The underlying physical, chemical, and biological mechanisms that govern the positive selectivity observed between the normal and cancerous tissue response in MRT are yet to be completely understood.…”

“…Laser micro-irradiation is a widely used approach having led to meaningful findings in cell biology, aging and cancer research 7,8,18,19 . With all its advantages over current existing manual evaluation tools, the Stripenator provides value and utility in objective, fast analyses and enables superior quality, high-throughput studies of migration and accumulation of DNA damage response factors, histone modifications, and DNA repair-related proteins.…”

“…The first microbeam device was used by Zirkle and Bloom, and consisted of a 2 MV Van de Graaff accelerator that delivered protons (15). Today, new-generation microbeam devices allow reducing the radiation beam to sub-cellular dimensions using collimation assemblies and electromagnetic focusing (16). External alpha particle microbeam irradiation has been and is a very attractive tool for exploring the radiobiology of alpha particles at the subcellular scale (17), although different from TAT in terms of the used dose and dose rate, and the absence of vector (18,19).…”
