Microarray analysis reveals global modulation of endogenous retroelement transcription by microbes

Abstract: BackgroundA substantial proportion of both the mouse and human genomes comprise of endogenous retroelements (REs), which include endogenous retroviruses. Over evolutionary time, REs accumulate inactivating mutations or deletions and thus lose the ability to replicate. Additionally, REs can be transcriptionally repressed by dedicated mechanisms of the host. Nevertheless, many of them still possess and express intact open reading frames, and their transcriptional activity has been associated with many physiologi… Show more

“…Genetic and/or epigenetic modulation of HERV elements at a particularly vulnerable development stage may lead to variable patterns of neurodevelopment and upon exposure to subsequent environmental triggers, such patterns may translate to a schizophrenia phenotype. Microbes that lead to immune cell activation through transcription of immune-related host genes may cause global modulation of endogenous retroelement transcription (Young et al, 2014). Certain copies of the HERV elements are activated by some infectious agents.…”

“…Interestingly, silenced endogenous sequences can be activated by innate immune responses, suggesting a role for these sequences in signal amplification. For instance, exposure to environmental microbes causes global modulation of endogenous retroelement transcription [71]. Moreover, treatment with IFN-I induces transcription of the short interspersed element (SINE), Alu, which in turn stimulates the secretion of proinflammatory cytokines in a TLR7-dependent manner.…”

“…We therefore hypothesize that HERV, like latent viruses, may reactivate and be transcribed in vivo after inflammatory injuries. Given that several groups showed that some probes of commercial whole genome microarray do target HERV loci (Young, Mavrommatis, and Kassiotis 2014; Reichmann et al 2012) (such as Affymetrix U133 plus 2), we retrospectively explored microarray datasets obtained in our lab to study the HERV transcriptome modulation in various contexts of injuries in vivo .…”
