Microanalytical systems for separations of stratum corneum ceramides

Abstract: Original PaperMicroanalytical systems for separations of stratum corneum ceramidesThe small amount of lipids from human skin obtained with noninvasive sampling method led us to investigate microanalytical separation techniques. The lipid class analysis was performed with a micro polyvinyl alcohol-silica (PVA-Silm) column. The gradient elution was from heptane to acetone/butanol 90:10 v/v in 4%/min at 78 lL/ min. In addition an evaporative light scattering detector (ELSD) was modified for micro-LC. All solvents… Show more

“…To analyze each individual synthetic CER with uniform chain length, NPLC was chosen over reverse phase LC because this mode allows for separation of individual CER subclasses. We chose to use a PVA column as it has advantages in terms of separation and peak shape compared with commonly used silica and diol columns ( 50,51 ). Although ESI is mainly used for the analysis of CERs, the APCI mode is also used frequently.…”

“…4 It demonstrated a highly hydrophilic property and has found some applications in super uid chromatography, 21 NPLC, 22 and HILIC. 23 It should be noted that the preparation of commercial PVA stationary phases was based on chemical bonding 21,22,24 and an obvious disadvantage of such a method is complicated procedures and lengthy preparation processes.…”

“…CLC offers interesting possibilities for the determination of lipids, as these compounds are not very sensitive in other instrumental separation techniques. An interesting application has been developed by L. Quinton et al [31]. ELSD can detect all solutes that are less volatile than the mobile phase.…”
