Mice lacking the proton channel Hv1 exhibit sex-specific differences in glucose homeostasis

Abstract: Sex as a physiologic factor has a strong association with the features of metabolic syndrome. Our previous study showed that loss of the voltage-gated proton channel Hv1 inhibits insulin secretion and leads to hyperglycemia and glucose intolerance in male mice. However, there are significant differences in blood glucose between male and female Hv1-knockout (KO) mice. Here, we investigated the differences in glucose metabolism and insulin sensitivity between male and female KO mice and how sex steroids contribu… Show more

“…Female mice had lower glucose at fasting and throughout GTT compared to male mice ( Figure 1 ). This is consistent with some past work ( 9 , 10 , 23 ) but not all ( 24 ). We show that LPS administration improved glucose tolerance in all mice, across sex and genotype ( Figure 2 ).…”

“…In humans, a higher hHv1 expression/function has been reported in several types of cancer cells such as breast cancer, , colorectal cancer, glioblastoma, and B-cell leukemia, whereas a loss of function seems to be involved in male infertility. On the other hand, working on the Hv1 KO mice, there is evidence that Hv1 inhibition protects the neuroinflammation induced by cerebral ischemic stroke , and induces several alterations in glucose metabolism. , …”

“…The control of the intracellular pH in the β-cells and its proper regulation is essential in the secretory machinery of insulin, which is the pivotal hormone in glucose homeostasis in mammals. It has been shown that Hv1 KO mice possess an impaired metabolism phenotype characterized by hyperglycemia and glucose intolerance due to a reduction in insulin secretion by the beta cells ( Pang et al, 2020 ; Pang et al, 2021 ). PMA stimulation (5 µM) of islet β-cells evoked a secretion of 2.5 ng of insulin per islet, while for the KO hvcn1 β-cells PMA induced a secretion 2.5-fold lower insulin per islet, 1 ng of insulin per islet.…”
