MHC Heterozygote Advantage in Simian Immunodeficiency Virus–Infected Mauritian Cynomolgus Macaques

Abstract: The importance of a broad CD8-T lymphocyte (CD8-TL) immune response to HIV is unknown. Ex vivo measurements of immunological activity directed at a limited number of defined epitopes provide an incomplete portrait of the actual immune response. Here we examined viral loads in SIVinfected MHC homozygous and heterozygous Mauritian cynomolgus macaques (MCM). Chronic viremia in MHC homozygous macaques was 80-fold greater than in MHC heterozygous macaques. Virus from MHC homozygous macaques accumulated 11 to 14 var… Show more

“…Sixteen overlapping PCR amplicons spanning the entire SIVmac239 genome were then amplified using a Qiagen OneStep reverse transcription (RT)-PCR kit (Qiagen). The conditions for RT-PCR were as described previously (37). Products were then gel purified and bidirectionally sequenced with a DYEnamic energy transfer terminator cycle sequencing kit (GE Healthcare, Piscataway, NJ) on an ABI 3730xl instrument (Applied Biosystems, Foster City, CA).…”

“…All animals were used in previous studies (9,11,12). They were purchased from Charles River Laboratories or Bioculture Group, and cared for by the Wisconsin National Primate Research Center (WNPRC) according to protocols approved by the University of Wisconsin Graduate School Animal Care and Use Committee.…”

“…These results indicate the association of MHC-I haplotypes with AIDS progression. There has been a number of reports describing SIV infections in macaques sharing a single or a couple of MHC-I alleles, but few studies have examined SIV infection in macaques sharing an MHC-I haplotype (10,11,40). SIV infection induces multiple epitope-specific CD8 ϩ T-cell responses, and CD8 ϩ T-cell responses specific for some MHC-I-restricted epitopes can be affected by those specific for other MHC-I-restricted epitopes due to CTL immunodominance (16,29,52).…”
