Methylene blue prevents retinal damage in an experimental model of ischemic proliferative retinopathy

Abstract: -Perinatal asphyxia induces retinal lesions, generating ischemic proliferative retinopathy, which may result in blindness. Previously, we showed that the nitrergic system was involved in the physiopathology of perinatal asphyxia. Here we analyze the application of methylene blue, a well-known soluble guanylate cyclase inhibitor, as a therapeutic strategy to prevent retinopathy. Male rats (n ϭ 28 per group) were treated in different ways: 1) control group comprised born-to-term animals; 2) methylene blue group … Show more

“…Sections were stained with hematoxylin and eosine. The number of RGCs per 25x objective microscopic field was estimated in each experimental group, as previously reported 54 . Additional retinas, taken from animals sacrificed 6 days after surgery, were stained for terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) with the In Situ Cell Death Detection Kit, POD (Roche, Basel, Switzerland), following manufacturer´s instructions.…”

“…For instance, it has been widely reported that it is a relatively potent inhibitor of SCD (IC 50 = 0.9 uM). However, SA is also able to mediate anti-inflammatory and protective effects in the retina independently of SCD (AP, RP, APS, IML, in preparation) by a mechanism that could also involve the toll-like receptor 4 (TLR4) receptor and intracellular kinases [58] and could be of interest in other retinal pathologies [120,121].…”

“…The changes observed in the retina of asphyctic animals include ganglion cell death, neovascularization, and Müller cell hypertrophy in the inner retina (Rey-Funes et al, 2013). The nitrergic system is one of the factors involved in the genesis of all these lesions (Rey-Funes et al, 2011;Rey-Funes et al, 2016).…”
