Methylation of DACT2 Promotes Papillary Thyroid Cancer Metastasis by Activating Wnt Signaling

Abstract: Thyroid cancer is the most common endocrine malignant disease and the incidence is increasing. DACT2 was found frequently methylated in human lung cancer and hepatocellular carcinoma. To explore the epigenetic change and the role of DACT2 in thyroid cancer, 7 thyroid cancer cell lines, 10 cases of non-cancerous thyroid tissue samples and 99 cases of primary thyroid cancer samples were involved in this study. DACT2 was expressed and unmethylated in K1, SW579, FTC-133, TT, W3 and 8505C cell lines. Loss of expres… Show more

“…In addition to deletion, epigenetic modifications of DACT genes were also reported. For instance, DACT1 and DACT2 were reported to be methylated in hepatocellular carcinoma, oral squamous, gastric cancer, nasopharyngeal carcinoma, thyroid cancer, colon cancer and lung cancer (10,(26)(27)(28)(29)(30)(31). In addition, DACT3 was identified to have histone modifications in colorectal cancer (25,26,32).…”

“…Along with these genetic factors, epigenetic regulation plays a crucial role in the initiation and progression of various types of cancers 20. Epigenetic alterations, such as aberrant promoter methylation, can yield powerful biomarkers for early detection of diseases 21, 22, 23. A previous research of Belinsky et al24 displayed that aberrant methylation of the p16 might be involved in hepatocellular cancer, which might be a novel biomarker for early detection.…”

“…Гіперметилювання REC8 пов'язано з поганим прогнозом у карциномах ЩЗ, включаючи їх прогресію та смерть пацієнта [63]. Ген, що кодує DACT (Dishevelled-associated antagonist of β-catenin), інгібітор сигнального каскаду WNT, пригнічується в РТС шляхом метилювання промотора [64]. Тканинний інгібітор металопротеїнази (TIMP) інгібує інвазійність, ангіогенез і метастазування.…”
