Methylated, blocked 5 termini in HeLa cell mRNA.

Abstract: Poly(A)-containing HeLa cell mRNA prepared from cells labeled with [3Plphosphate was found to contain a variety of methylated, blocked 5'-terminal structures of two general types: m7GpppNm-Np and m7GpppNm-Nm-Np. In addition, about one-third of the [3Hlmethyl label was present in the N6-methyladenosine; this labeled nucleoside was not found in the 3'-terminal one-third of the mRNA chain and thus may also be in the 5' portion of the mRNA.The 5' end of a variety of viral mRNA molecules consists of an unusual typ… Show more

“…That same structure was soon found also in both cellular nuclear RNA and polyribosomal mRNA (Perry and Kelley 1974;Furuichi et al 1975b;Wei et al 1975;Salditt-Georgieff et al 1976). …”

“…One possible modification between the first and second bases of an mRNA would be a 29-O-methylation of the sugar attached to the first templated base of the splice leader. This modification has been observed in other systems (e.g., Furuichi et al 1975) and would certainly be expected to influence the rate of alkali cleavage.…”

“…The presence of a 5= cap structure m 7 G(5=)ppp(5=)Nm is a general feature of eukaryotic mRNAs and many viral mRNAs that provides protection from exonuclease digestion and enhances translation (1)(2)(3)(4)(5)(6). In eukaryotic cells, mRNA decay begins with shortening of the poly(A) tail and proceeds in either the 5=-to-3= or 3=-to-5= direction.…”
