Methods for the Cost-Effective Production of Bacteria-Derived Double-Stranded RNA for in vitro Knockdown Studies

Abstract: RNA interference (RNAi) is a highly conserved pathway for the post-transcriptional regulation of gene expression. It has become a crucial tool in life science research, with promising potential for pest-management applications. To induce an RNAi response, long double-stranded RNA (dsRNA) sequences specific to the target gene must be delivered to the cells. This dsRNA substrate is then processed to small RNA (sRNA) fragments that direct the silencing response. A major obstacle to applying this technique is the … Show more

“…Moreover, the application of living dsRNA-producing bacteria for gene silencing has been used in nematodes [ 160 ], insects [ 225 ], crustaceans [ 226 ], mammalian cells [ 227 ], and filamentous fungi [ 215 ]. However, in order to avoid bacterial release into the environment, it is necessary to purify the RNA, which can make the process more expensive and limit the amount of dsRNAs produced [ 26 , 228 , 229 , 230 , 231 , 232 ].…”

“…Production of a large amount of dsRNA is required for silencing the expression of a gene in vivo. The bacterial system is the most common between in vivo dsRNA expressing alternatives [136]. This production involves the efficient release of the dsRNA from the cells without affecting RNA integrity during the extraction process [136].…”

“…The bacterial system is the most common between in vivo dsRNA expressing alternatives [136]. This production involves the efficient release of the dsRNA from the cells without affecting RNA integrity during the extraction process [136]. In experimental settings three main approaches are used to produce dsRNA: chemical synthesis of NTPs, in vitro synthesis through RNA-dependent RNA polymerases and fermentation through microorganisms.…”

“…(Goodfellow et al The application of dsRNA insecticides via direct spraying of the microbial fermentation material (expressing the dsRNA) has potential drawbacks related to crop spraying itself as well as challenges and costs of the formulation. Although this effectively triggers knockdown of some insects' target genes including, hemocoel of orthopteran insects (Verdonckt & Vanden Broeck, 2022), it is less effective in insects such as Helicoverpa pests due to immune responses to the presence of bacteria in the hemolymph (Li et al, 2019).…”
