Methodology of Pulsed Photoacoustics and Its Application to Probe Photosystems and Receptors

Abstract: We review recent advances in the methodology of pulsed time-resolved photoacoustics and its application to studies of photosynthetic reaction centers and membrane receptors such as the G protein-coupled receptor rhodopsin. The experimental parameters accessible to photoacoustics include molecular volume change and photoreaction enthalpy change. Light-driven volume change secondary to protein conformational changes or electrostriction is directly related to the photoreaction and thus can be a useful measurement… Show more

“…The evaluation of light energy storage (ES) was performed in accordance with the generally accepted principles and practical approaches 5 6 7 30 using the previously described fast Fourier transform (FFT) photoacoustic method 13 with modifications. Leaf discs (10 mm in diametre) were cut from the peripheral part of the corollas where the residual chlorophyll content is minimal.…”

“…Such an increase occurs only when the photobaric component is eliminated57. Considering the kinetic and spectral parametres of the PA-signal, a nonzero ES value provides an incontrovertible and direct evidence of either oxygenic or anoxygenic photosynthesis567.…”

“…Far-red light sources are widely used in photoacoustic studies of biochemical processes involving CET-PSI (Joliot et al, 2006;Hou and Sakmar, 2010). Besides, far-red light can be used in dualwavelength excitation experiments to test PSI and PSII coupling within a single linear electron transport chain (Lysenko, 2012).…”

“…Pulsed photoacoustic spectroscopy can provide a direct measurement of thermodynamic parameters such as volume change and enthalpy changes that accompany electron transfer reactions (Braslavsky, 1985;Carpentier et al, 1990;Arnaut et al, 1992;Small et al, 1992;Losi et al, 1995;Edens et al, 2000;Malkin, 2000;Herbert et al, 2001;Feitelson and Mauzerall, 2002;Delosme, 2003;Hou and Mauzerall, 2006;Hou and Sakmar, 2010;Hou and Mauzerall, 2011). With prior knowledge of the change in Gibbs free energy of the corresponding reactions, the apparent entropy change (TS) of the reaction can be calculated.…”
