Methodology of predicting novel key regulators in ovarian cancer network: a network theoretical approach

Abstract: BackgroundIdentification of key regulator/s in ovarian cancer (OC) network is important for potential drug target and prevention from this cancer. This study proposes a method to identify the key regulators of this network and their importance.MethodsThe protein-protein interaction (PPI) network of ovarian cancer (OC) is constructed from curated 6 hundred genes from standard six important ovarian cancer databases (some of the genes are experimentally verified). We proposed a method to identify key regulators (… Show more

“…Thus, the important role of the PI3K/protein kinase B (AKT)/mammalian target of rapamycin (mTOR) and NF-κB pathways is emphasized, as in OvCa they are often hyperactive and susceptible to mutations, which correlates with a poor prognosis [ 36 ]. Bioinformatics methods have revealed five key regulators in OvCa: AKT1 , KRAS , EPCAM , CD44 , and MCAM ; AKT1 , as one might expect, plays the most important role [ 37 ]. A separate significant point is the ratio of AKT pathways and autophagy.…”

“…Removal of high degree nodes (hubs) in biological networks may cause lethality in various complex biological networks. This idea of understanding may enable us to determine the topological features of the network architecture, important functionally related modules and hubs [55,56]. The topological properties were calculated after each subsequent removal using the Network Analyzer plugin in Cytoscape version 3.7.1.…”

“…The PPI study of the identified protein sets and the protein community finding by in silico network analysis identified a set of key regulator molecules [42][43][44][45] Upon TXR treatment, the C9, AAG and catalase abundance were decreased almost to the basal levels without significant alteration in CRP, a non-specific indicator of inflammation. High aldehyde dehydrogenase (ALDH) activity has been reported in osteoarthritis patients'…”
