Methionine restriction fundamentally supports health by tightening epithelial barriers

Abstract: Dietary methionine restriction (MR) has been found to affect one of the most primary tissue-level functions of an organism: the efficiency with which the epithelial linings of major organs separate the fluid compartments that they border. This process, epithelial barrier function, is basic for proper function of all organs, including the lung, liver, gastrointestinal tract, reproductive tract, blood-brain barrier, and kidney. Specifically, MR has been found to modify the protein composition of tight junctional… Show more

“…Shifts in intestinal microbiota profiles may cause intestinal inflammation and affect the expression of tight junction-related proteins leading to increased intestinal permeability [ 18 , 33 , 69 ]. Furthermore, modulation of the methionine dietary intake has recently been reported to affect the expression of the intestinal tight junction-related proteins [ 56 ]. Gene expression analysis revealed a non-significant trend towards increase (2.6-fold, p < 0.2) in the expression of Tnfα in the proximal jejunum of mice fed MSD, compared to MAD-fed mice on day 6.…”

“…Another role of AMPK in TJ formation and maintenance could be hypothetically linked to dietary methionine restriction (MR). MR has been found to modify the protein composition of TJ complexes in epithelial cells [ 125 ]. In addition, the stimulation of S- adenosyl-l-methionine, a key intermediate of methionine metabolism, led to the consumption of both Met and ATP [ 126 ] and AMPK activation [ 127 ].…”

“…However, methionine restriction to a level of 0.12-0.15% in rodent diets (compared to 0.60-0.86% in a standard diet, and similar to a plant-based diet [12]) is consistently associated with an increase in lifespan in animal models [13][14][15][16][17]. It is also associated with many health benefits, such as improvements in insulin and lipid regulation and barrier function [18]. Furthermore, methionine restriction significantly impairs tumor growth and metastasis in preclinical models [10,19].…”
