Rheumatoid arthritis (RA) is a debilitating disease associated with locomotion impairment, and conventional therapeutic drugs are not optimal for managing RA. There is an avalanche of medications used for the management of RA. Still, studies have shown that they are associated with severe side effects, including hepatotoxicity, retinopathy, and cardiotoxicity disorders of the central nervous system (CNS), skin, blood, and infections. Complementary and alternative medicine (CAM) is currently gaining attention as a novel panacea for managing debilitating diseases, such as RA. Nigerian folk herbal remedies are replete with a plethora of curative medicine, albeit unvalidated scientifically but with seemingly miraculous provenance. Studies of the identification of bioactive compounds present in these botanicals using advanced spectral analytical techniques have enhanced our understanding of the role of Nigerian herbal remedies in the treatment and management of RA. Interestingly, experimental studies abound that the bioactive compounds present in the extracts of plant botanicals protected animals from the development of RA in different experimental models and reduced the toxicity associated with conventional therapeutics. Validated mechanisms of RA amelioration in human and animal models include suppression of the expression of NF-κB, IL-1β, TNF-α, IL-6, IL-8, IL-17, IL-23, chemokines, TGF-β, RANKL, RANK, iNOS, arginase, COX-2, VEGFA, VEGFR, NFATC1, and TRAP in the synoviocytes. Decreased ROS, NO, MDA, carbonyl groups, and PGE2 in the synovial fluid increased the expression of PPARα/γ; antioxidant and anti-inflammatory molecules also improve RA etiology. In this mini-review, we discuss the global burden of RA, the novel role of plant-based botanicals as potential therapeutics against signaling pathways in RA. Also addressed is the possible repurposing/reprofiling of plant botanicals to increase their therapeutic index among RA patients that patronize traditional healers in Nigeria with a global projection.

The purpose of this mini-review was to summarize and update knowledge on the phytochemistry, pharmacology, and toxicity of Cola nitida, with the view of providing baseline data for herbal drug formulation. In January 2021, a non-exhaustive online search of relevant articles was carried out on the phytochemistry, pharmacology, and toxicology of C. nitida from scientifically well-established databases such as Science Direct, PubMed, Web of Science, Scopus, Google Scholar, and SciELO. The plant's scientific name as well as phytochemistry, pharmacology, pharmacognosy, bioactivity and toxicology were used as keywords. The chemical structures of the compounds isolated from this plant were drawn using ChemBioDraw Ultra 12.0 software. A literature survey has revealed that C. nitida is highly appreciated by African populations in various cultures, especially in West Africa. Phytochemical analyses showed that C. nitida contains interesting compounds like catechin, caffeine, epicatechin, polyphenols, alkaloids, tannins, saponins, bromelain, cardenolides, proanthocyanidins, triterpenes, glycosides, flavonoids, anthraquinones, steroids, anthocyanins, glycosides, alkaloids, etc. The presence of these phyto-compounds in the investigated plant species justifies its used as an antimicrobial, anti-malarial, anti-inflammatory, anti-diabetic, anti-coagulant agent. Thus, C. nitida could be used as a raw material for manufacturing efficient medication against various diseases, including sickle cell disease.

Background Medicinal plants are used to manage pain and inflammatory disorders in traditional medicine. A scientific investigation could serve as a basis for the determination of molecular mechanisms of antinociceptive and antiinflammatory actions of herbal products. In this work, we used both artificial intelligence (AI) based models inform of adaptive neuro-fuzzy inference system and artificial neural network (ANN) as well as a linear model, namely; stepwise linear regression in modelling the performance of four different inflammatory biomarkers namely; interleukin (1L)-1β, 1L-6, tumour necrosis factor (TNF)-α and prostaglandin E2 (PGE2). This modelling was done using number of abdominal writes, the reaction time of paw licking in mice and paw oedema diameter as the input variables. Results Four different performance indices were employed, which are determination coefficient (DC), root mean squared error (RMSE), mean square error (MSE) and correlation co-efficient (CC). The results have shown the superiority of the AI-based models over the linear model. Conclusions The overall quantitative and visualized comparison of the results showed that adaptive neuro-fuzzy inference system outperformed the ANN and SWLR models in modelling the performance of the four inflammation biomarkers in both the calibration and verification phases.
