Metformin Attenuates Cyclosporine A-induced Renal Fibrosis in Rats

Abstract: Background. The aim of the present study was to investigate the therapeutic potential of metformin in preventing cyclosporine A (CsA)-induced nephrotoxicity. Methods. Three groups of adult male Sprague-Dawley rats were treated with vehicle, CsA, and CsA + metformin for 4 weeks following 1 week on low sodium diet, respectively. At the end of treatment, all animals were euthanized, and the samples of kidney, urine, and blood were collected for functional,… Show more

“…Additionally, metformin treatment has been demonstrated to slow cystogenesis in a murine model of autosomal-dominant polycystic kidney disease [ 30 ]. It has also been reported to reduce renal fibrosis in high-fat diet-fed rats [ 31 ], unilateral ureteral obstruction (UUO) mice [ 32 , 33 ], folic acid-induced renal fibrosis [ 32 , 34 ], adenine-induced chronic kidney disease rats [ 35 ], and cyclosporine A-induced renal fibrosis rats [ 36 ]. Moreover, accumulating evidence has shown that metformin has great potential in alleviating the progression of fibrosis in various organs.…”

“…As a commonly used clinical immunosuppressant, cyclosporine A (CsA) has been widely prescribed for inhibiting rejection after organ transplantation. Many experimental studies showed that long-term use of CsA could contribute to progressive renal interstitial fibrosis, renal cell apoptosis, and immune cell infiltration ( Wu et al, 2018 ; Lin et al, 2019 ). Findings from Doh’s group showed that Panax ginseng C.A.Mey.…”
