Metallothionein-III Prevents γ-Ray-induced 8-Oxoguanine Accumulation in Normal and hOGG1-depleted Cells

Abstract: Metallothioneins (MT) play an important biological role in preventing oxidative damage to cells. We have previously demonstrated that the efficiency of the protective effect of MT-III against the DNA degradation from oxidative damage was much higher than that of MT-I/II. As an extension of the latter investigation, this study aimed to assess the ability of MT-III to suppress 8-oxoguanine (8-oxoG), which is one of the major base lesions formed after an oxidative attack to DNA and the mutant frequency of the HPR… Show more

“…One of the protective mechanisms attributed to cadmium involves induction of metallothioneins, which are reported to be involved in protection against chemically induced oxidative stress and also act as a free radical scavenger (Jeong et al 2004;Takahashi et al 2004). DeMicco et al (2010) observed that treatment with diazepam ameliorated the spasms while treatment with the sodium channel antagonist MS-222 ameliorated both spasms and body curvature which were reminiscent of the classic syndromes observed with pyrethroid toxicity in zebrafish.…”

“…Recent studies suggest that MT3 is involved in the protection of reactive oxygen species-induced DNA damage [28]. MT3 prevents the gamma-radiation-induced 8-oxoG accumulation and mutation in normal and hOGG1-depleted cells [29]. Because of MT3s antioxidant functions, we investigated the expression of MT3 and its epigenetic regulation in esophageal adenocarcinoma.…”

“…The 8-oxoG level in the DNA is expressed as 8-oxoG/10 7 dG. HPRT Mutation Assay-The method used to determine the frequency of the ␥-ray-induced HPRT mutants has been described (57). Briefly, cells were plated at 5 ϫ 10 4 /100-mm diameter dish and cultured in Earle's minimal essential medium, 10% fetal bovine serum, 100 M hypoxanthine, 0.4 M aminopterin, and 16 M thymidine (Invitrogen) for at least 14 days.…”
