As a new class of agricultural insecticides, synthetic pyrethroids are widely used to control insect pests. Synthetic pyrethroids have been shown to enter the aquatic environment from agricultural runoff or drift from aerial and ground-based spraying applications posing threat to fishes which are less tolerant to pesticides through direct exposure. These insecticides interfere with the sodium channel of the nervous system resulting in prolonged sodium tail current. Widespread application of these chemicals has warranted the attention of the ecologist to understand the impact of these chemicals on the aquatic environment. In this perspective, an updated account of toxicological evaluation of three type II synthetic pyrethroids, viz. deltamethrin, cypermethrin and fenvalerate in terms of their physico-chemical, metabolic, hematological, histological, behavioral and reproductive aspects with respect to the fishes has been presented which may be useful for policy makers, academics, environmental scientists and agricultural professionals needing ready access to this information. The aim of the present synoptic literature appraisal was to summarize the main effect of current use, type II synthetic pyrethroids (deltamethrin, cypermethrin and fenvalerate) on aquatic environment due to their persistence and accumulation. This article will focus on non-target organisms in inland fresh water environment with special reference to fin fishes and will critically evaluate the toxicity of these pyrethroids in terms of growth inhibition, metabolic disorders, neurotoxicity, reproductive failure, enzymatic dysfunction, haematological alterations, and tissue damages. The rationalized information in this milieu may be useful in ecological risk evaluation and human health management as fish serves as an important bio-indicator for aquatic systems health.

BackgroundMetallothionein 3 (MT3) maintains intracellular metal homeostasis and protects against reactive oxygen species (ROS)-induced DNA damage. In this study, we investigated the epigenetic alterations and gene expression of the MT3 gene in esophageal adenocarcinomas (EACs).Methods and ResultsUsing quantitative bisulfite pyrosequencing, we detected unique DNA methylation profiles in the MT3 promoter region. The CpG nucleotides from −372 to −306 from the transcription start site (TSS) were highly methylated in tumor (n = 64) and normal samples (n = 51), whereas CpG nucleotides closest to the TSS (−4 and +3) remained unmethylated in all normal and most tumor samples. Conversely, CpG nucleotides in two regions (from −139 to −49 and +296 to +344) were significantly hypermethylated in EACs as compared to normal samples [FDR<0.001, −log10(FDR)>3.0]. The DNA methylation levels from −127 to −8 CpG sites showed the strongest correlation with MT3 gene expression (r = −0.4, P<0.0001). Moreover, the DNA hypermethylation from −127 to −8 CpG sites significantly correlated with advanced tumor stages and lymph node metastasis (P = 0.005 and P = 0.0313, respectively). The ChIP analysis demonstrated a more repressive histone modification (H3K9me2) and less active histone modifications (H3K4me2, H3K9ace) in OE33 cells than in FLO-1 cells; concordant with the presence of higher DNA methylation levels and silencing of MT3 expression in OE33 as compared to FLO-1 cells. Treatment of OE33 cells with 5-Aza-deoxycitidine restored MT3 expression with demethylation of its promoter region and reversal of the histone modifications towards active histone marks.ConclusionIn summary, EACs are characterized by frequent epigenetic silencing of MT3. The choice of specific regions in the CpG island is a critical step in determining the functional role and prognostic value of DNA methylation in cancer cells.

Cadmium is a well known human and animal carcinogen and is a ubiquitous contaminant in the environment. Although the carcinogenic mechanism of cadmium is a multifactorial process, oxidative DNA damage is believed to be of prime importance. In particular, cadmium suppresses the capacity of cells to repair oxidative DNA damage. In this study, cadmium treatment led to a significant increase in ␥-ray-induced 8-oxoguanine (8-oxoG) formation. Western blotting and semiquantitative reverse transcription-PCR revealed that cadmium treatment caused a decrease in the expression level of human OGG1 (8-oxoguanine-DNA glycosylase-1; hOGG1) in human fibroblast GM00637 and HeLa S3 cells. In addition, the cadmium-mediated decrease in hOGG1 transcription was the result of decreased binding of the transcription factor Sp1 to the hOGG1 promoter. Finally, we show that an increase in the functional hOGG1 expression level could inhibit the cadmium-mediated increase in ␥-ray-induced 8-oxoG accumulation as well as in ␥-radiation-induced mutation frequency at the HPRT (hypoxanthine-guanine phosphoribosyltransferase) gene locus. These results suggest that cadmium attenuates removal of ␥-ray-induced 8-oxoG adducts, which in turn increases the mutation frequency, and that this effect might, at least in part, result from suppression of hOGG1 transcription via inactivation of Sp1 as a result of cadmium treatment.
