Metabonomics: a platform for studying drug toxicity and gene function

Abstract: The later that a molecule or molecular class is lost from the drug development pipeline, the higher the financial cost. Minimizing attrition is therefore one of the most important aims of a pharmaceutical discovery programme. Novel technologies that increase the probability of making the right choice early save resources, and promote safety, efficacy and profitability. Metabonomics is a systems approach for studying in vivo metabolic profiles, which promises to provide information on drug toxicity, disease pro… Show more

“…Hydrazine also exhibited a lower AUC value than the other data sets, indicating poorer discrimination. One reason for these observations is the strong variability in speed of response of individual animals to the toxic insult in this case (so-called fast and slow responders 19 ). For some metabolites, this produced a wide range of intensities at the single time point examined, resulting in both closure-induced correlations and high biological correlations, thus increasing the overlap between distributions.…”

“…The measurement of metabolome variations in response to stimuli has been widely used in modern pharmaceutical and biomedical research [1,2]. The main analytical methods employed for metabonomic studies are based on informationrich spectroscopic techniques, such as nuclear magnetic resonance (NMR) spectrometry and mass spectrometry (MS), which is usually coupled with gas chromatography (GC/MS), liquid chromatography (LC/MS) or capillary electrophoresis (CE/MS) [3][4][5].…”

“…This method has recently demonstrated enormous potentials in many fields such as plant genotype discrimination [2][3], toxicological mechanisms, disease processes, and drug discovery [4][5][6][7][8][9][10]. One such recent application of this method included the rapid and noninvasive diagnosis of coronary heart disease [11][12][13][14]. In these methods, metabolite profiling is mainly used for the analysis of a class of metabolites.…”
