Metabolomics Reveals Metabolic Changes Caused by Low-Dose 4-Tert-Octylphenol in Mice Liver

Abstract: Background: Humans are constantly exposed to low concentrations of 4-tert-octylphenol (OP). However, studies investigating the effects of low-dose OP on the liver are scarce, and the mechanism of these effects has not been thoroughly elucidated to date. Methods: Adult male institute of cancer research (ICR) mice were exposed to low-dose OP (0, 0.01 and 1 μg/kg/day) for 7 consecutive days. Weights of mice were recorded daily during the experiment. Blood serum levels of OP, alanine aminotransferase (ALT) and asp… Show more

“…Uridine was also reported to exert protective effects against hepatotoxicity ( Zhao et al, 2018 ). Therefore, the reduction of uridine in our study may indicate that the DNA and RNA synthesis was disturbed in the host and liver toxicity was induced by the infection ( Zhou et al, 2018 ). It was reported that selenomethionine plays an important role in preventing oxidative stress and improving cell viability ( Bai et al, 2019 ).…”

“…Samples were prepared according to the previous literature [ 27 ]. The tissue preparation steps were as follows: 50 mg of frozen tissue was cut with surgical scissors, then mixed with 750 μL ultra-pure water and ultrasonicated (power: 60%).…”

“…The metabolomic analysis was performed as a previous report [ 27 ]. Briefly, UPLC Q-Exactive MS analysis was performed using a UPLC Ultimate 3000 system (Dionex, Germering, Germany) plus a Q-Exactive mass spectrometer (Thermo Fisher Scientific, Bremen, Germany) in both positive and negative modes with fullscan acquisition (70–1050 m/z ).…”

“…After drying, the residue was reconstituted for metabolomics analysis. The extracted samples were subjected to unbiased metabolomics analysis according to protocols in our previous studies 52,53 . Metabolomics analysis was performed on a UPLC Ultimate 3000 system (Dionex, Germering, Germany) coupled to a Q-Exactive Orbitrap mass spectrometer (Thermo Fisher Scientific, Bremen, Germany).…”
