Metabolomics as a tool for cardiac research

Abstract: Metabolomics represents a paradigm shift in metabolic research, away from approaches that focus on a limited number of enzymatic reactions or single pathways, to approaches that attempt to capture the complexity of metabolic networks. Additionally, the high-throughput nature of metabolomics makes it ideal to perform biomarker screens for diseases or follow drug efficacy. In this Review, we explore the role of metabolomics in gaining mechanistic insight into cardiac disease processes, and in the search for nove… Show more

“…Only one sample from the control set and one from the group of multiple myeloma patients at the time of diagnosis were identified as strong outliers on their group, outside the Hotellings 95% confidence limit. They displayed a metabolic profile that has been previously reported to be related to hypertensive patients (17,18), characterized by changes in signals attributed to lipids and lipoproteins, as well as the choline-containing region. Furthermore, the detailed revision of the clinical history of the multiple myeloma patient at the time of diagnosis revealed that the patient was diabetic and hypertensive.…”

“…Detailed information regarding NMR theory, its application and typical chemical shift values is available elsewhere (Bl眉mich 2005). The application of NMR spectroscopy in metabolomics ranges from pharmaceutical studies (Lindon et al 2007) to cardiovascular biomarker identification (Griffin et al 2011, Rhee & Gerszten 2012.…”

“…The metabolomic fingerprint, derived from specific protein enzymatic processes, directly correlates with the clinical phenotype of the diseases. [1][2][3] Cardiac resynchronization therapy (CRT) has shown a clear clinical benefit in heart failure patients by improving symptoms, quality of life, exercise capacity and cardiac function, significantly reducing morbidity and mortality, 4-9 but up to 40% of heart failure patients are nonresponder to CRT. 10,11 As an emerging discipline for molecular profiling, metabolomic may increase understanding of human diseases and clinical risk because changes in metabolite levels provide a real-time estimate of disease state and reflect the integrated effects of genomic, transcriptomic and proteomic variation.…”
