Metabolomics applications of FT‐ICR mass spectrometry

Abstract: Metabolomics, also known as Metabolic Profiling, is an emerging discipline under the umbrella concept of systems biology. The goal of metabolomics is to know and understand the concentrations and fluxes of endogenous metabolites within a living biological system under study. General tools are being developed for the rapid measurement of many metabolites in a single experiment, most of which are mass spectrometric methods. FT-ICR has unique advantages, as a mass spectrometric method, in this regard. Application… Show more

“…Upon performing internal calibration, we obtained an average accuracy below 1 ppm in agreement with previous reports using similar equipment. 26,27 We compared all samples according to the criteria mentioned in the Material and Methods (Selection of Differentially Expressed Peptides) and identified 422 peptide masses as significantly differentially expressed. Correlation analysis based on measured intensities of these 422 peptide masses is shown in Figure 3.…”

“…The FT-ICR-MS instrument is one of great future potential since it currently off ers the highest levels of resolution (>1000000 for non-complex samples) and mass precision (typically within 1-2 ppm). The instrument's dynamic range is also good (∼5000) (Barrow et al, 2005;Brown et al, 2005). However, due to the low scan speeds employed by FT-ICR-MS, typically 1 scan event per second, it is more appropriate for the direct infusion of samples rather than to apply a priori LC.…”

“…Mass spectrometry has long been the favourite method for comprehensive metabolome screening; it has the largest scope for metabolite analysis, in terms of the universality, sensitivity and specificity of the measurements 19,22 . Several mass-spectrometric platforms can measure large numbers of metabolites simultaneously in any given sample 18,[23][24][25][26] . ultra-high-accuracy mass analysers 27 achieve molecular-mass resolutions of up to 1,000,000 and mass accuracies that are better than 1 part per million.…”
