Metabolomics analysis reveals aminoquinazolin derivative 9d-induced oxidative stress and cell cycle arrest in A549 cells

Abstract: A novel aminoquinazolin derivative (named 9d), synthesized in our lab shows potential antitumor activity against A549 lung cancer cell lines. However, previous studies on the pharmacological mechanism of 9d mostly focused on cell and gene levels; the metabolic mechanism remains unknown. In this paper, an ultra-performance liquid chromatography coupled to quadrupole time-of-flight mass spectrometry (UPLC/Q-TOF MS) based metabolomics approach was established to study the probable antitumor mechanism. Q-TOF MS an… Show more

“…In non-infected cells, the cell cycle distribution in DMSO-treated A549 cells at 24 h.p.i was 73% in the G0/G1 phase, 10% in the S phase and 17% in the G2/M phase, as expected 31 , 32 . Treatment with SINE compounds significantly altered the distribution of cells in each phase of the cell cycle (Fig.…”

“…Hence, it was speculated that aminoacyl-tRNA biosynthesis was inhibited. Aminoacyl-tRNA biosynthesis is occurring at the G1 phase, as catalyzed by tRNA synthase [ 15 ]. When tRNA synthase is inhibited, the aminoacyl-tRNA biosynthesis is blocked and cells will arrest in the G1 phase.…”

“…Arginine is directly related to the production of nitric oxide in the body and participates in the regulation of end-stage renal degeneration [18-20]. In addition, the aminoacyl-tRNA biosynthesis pathway is related to oxidative stress, cell-cycle arrest, and apoptosis [21]. Besides arginine, several core differential metabolites are worth discussing.…”
