Metabolomic and proteomic characterization of sng and pain phenotypes in fibromyalgia

Abstract: Background: Fibromyalgia (FM) is characterized by chronic widespread pain. Its pathophysiological mechanisms remain poorly understood, and effective diagnosis and treatments are lacking. This study aimed to identify significantly changed biosignatures in FM and propose a novel classification for FM based on pain and soreness (sng) symptoms. Methods: Urine and serum samples from 30 FM patients and 25 controls underwent metabolomic and proteomic profiling. Results: Compared with controls, FM patients showed signifi… Show more

“…Inter-α-trypsin inhibitor heavy chain H4 (ITIH4), a member of the serine protease inhibitor family, is a liver-originated plasma glycoprotein (Ma et al 2021;Zhuo and Kimata 2008). It has been reported that ITIH4 modulates atherosclerosis and lipids, which are involved in the development of AIS (Hao et al 2022;Hsu et al 2022;Malaud et al 2012). Importantly, ITIH4 has the capability of regulating inflammation, the latter is detrimental to the severity and functional outcomes of AIS (Huang 2022;Ma et al 2021;Shi et al 2019).…”

“…46 In addition, LRG is also a biomarker of fibromyalgia. 23,47,48 As serum IL-6 levels are elevated in subjects with fibromyalgia, it is suggested that fibromyalgia may affect the assessment of inflammatory activity in patients with RA. 49 This is in line with the results of this study, which showed that RA subjects with OA and fibromyalgia had the highest concentration of serum LRG.…”

“…However, the mechanism used by RNF11 to regulate protein ubiquitination remains unclear because the RNF11 promotes ubiquitination in some cases but inhibits substrate ubiquitination in other instances [ 31 ]. Hsu et al conducted a metabolomic and proteomic analysis of serum samples from patients with FM, and the results showed that FM patients had metabolic disorders and produced numerous NF- κ B-dependent cytokines [ 32 ]. RNF11 has been reported as a negative regulator of NF- κ B and Jun N-terminal kinase (JNK) signaling pathways [ 33 ].…”
