Metabolomic and genetic analyses of flavonol synthesis in Arabidopsis thaliana support the in vivo involvement of leucoanthocyanidin dioxygenase

Abstract: Flavonol synthase (FLS) (EC-number 1.14.11.23), the enzyme that catalyses the conversion of Xavonols into dihydroXavonols, is part of the Xavonoid biosynthesis pathway. In Arabidopsis thaliana, this activity is thought to be encoded by several loci. In addition to the FLAVONOL SYNTHASE1 (FLS1) locus that has been conWrmed by enzyme activity assays, loci displaying similarity of the deduced amino acid sequences to FLS1 have been identiWed. We studied the putative A. thaliana FLS gene family using a combination … Show more

“…Similar staining experiments performed in Arabidopsis identified yellow and green DPBA-mediated fluorescent compounds as quercetin and kaempferol (flavonols) and blue fluorescence-emitting compounds as sinapate glycoside derivatives (Stracke et al, 2007(Stracke et al, , 2009). Application of additional staining, including Shinoda's test and Zn-HCl (Pew's test) on yellow and white F 3 methanol extracts (Supplemental Fig.…”

“…Since no FLS like sequence was obtained during our cloning studies and the expression of related Xavonol/anthocyanin pathway genes such as FHT, DFR and ANS is rather low and speciWc compared to the transcripts of Xavone biosynthesis, the absence of Xavonols is likely. However, due to the described site activity of ANS (Stracke et al 2009) the synthesis of Xavonols cannot be excluded and they might escape detection due to low, tissue speciWc, or spatial occurrence. UGT90A7 showed highest activity with eriodictyol and luteolin.…”
