To investigate the efficacy of TP (Toripalimab Plus) chemotherapy combined with karelizumab in the treatment of advanced ovarian cancer after surgery. We recruited 96 patients with advanced ovarian cancer who were admitted to our hospital between December 2019 and December 2022 with advanced ovarian cancer and treated by tumor cell reduction surgery. The patients were randomly divided into groups using a digital table, 48 each. The control group was treated with chemotherapy, while the observation group was treated with chemotherapy combined with karelizumab. We compared the two groups with regards to adverse reactions and the levels of VEGF (Vascular Endothelial Growth Factor), MMP-9 (matrixmetalloproteinase 9), CEA (A Carcinoma Embryonic Antigen), AFP (alpha-fetoprotein), CA125 (carbohydrate antigen 125), CA19-9 (carbohydrate antigen 19-9) and T lymphocytes. There was no significant difference between the two groups with regards to adverse reactions (p > 0.05); the levels of VEGF and MMP-9 in the observation group were lower. There was a significant difference between the data before and after treatment within the same group (all p < 0.05); the levels of CEA, AFP, CA125 and CA19-9 in the observation group were lower than those in the control group, with significant differences before and after treatment within the same group (p < 0.05); the levels of CD3+ (cluster of differentiation 3+) and CD4+ in the observation group were higher, while the levels of CD8+ were lower than those in the control group. There were significant differences before and after treatment in the same group (p < 0.05). The combination of karelizumab and TP chemotherapy had a significant and positive impact on postoperative patients with advanced ovarian cancer by effectively regulating immune function and the levels of tumor markers. This protocol is safe and can be selected by considering the specific situation of individual patients.

Oncocytic thyroid cancer is characterized by the aberrant accumulation of abnormal mitochondria in the cytoplasm and a defect in oxidative phosphorylation. We performed metabolomics analysis to compare metabolic reprogramming among the oncocytic and non-oncocytic thyroid cancer cell lines XTC.UC1 and TPC1, respectively, and a normal thyroid cell line Nthy-ori 3-1. We found that although XTC.UC1 cells exhibit higher glucose uptake than TPC1 cells, the glycolytic intermediates are not only utilized to generate end-products of glycolysis, but also diverted to branching pathways such as lipid metabolism and the serine synthesis pathway. Glutamine is preferentially used to produce glutathione to reduce oxidative stress in XTC.UC1 cells, rather than to generate α-ketoglutarate for anaplerotic flux into the TCA cycle. Thus, growth, survival and redox homeostasis of XTC.UC1 cells rely more on both glucose and glutamine than do TPC1 cells. Furthermore, XTC.UC1 cells contained higher amounts of intracellular amino acids which is due to higher expression of the amino acid transporter ASCT2 and enhanced autophagy, thus providing the building blocks for macromolecules and energy production. These metabolic alterations are required for oncocytic cancer cells to compensate their defective mitochondrial function and to alleviate excess oxidative stress.

ObjectiveTumor cells can acquire a large amount of energy and structural components by reprogramming energy metabolism; moreover, metabolic profiles slightly differ according to cancer type. This study compared and assessed the metabolic profile of head and neck squamous cell carcinoma (HNSCC) and normal tissues, which were collected from patients without cancer.Subjects and MethodsOverall, 23 patients with HNSCC and 6 patients without cancer were included in the analysis. Metabolomic profiles were analyzed using capillary electrophoresis‐mass spectrometry. Gene expression was evaluated using real‐time reverse transcription‐polymerase chain reaction.ResultsGlycolysis, the pentose phosphate pathway, tricarboxylic acid cycle, and glutamine metabolism were upregulated in HNSCC tissues based on gene expression analysis. HNSCC could then have enhanced energy production and structural component. The levels of lactate, succinate, glutathione, 2‐hydroxyglutarate, and S‐adenosylmethionine, considered as oncometabolites, increased and these had accumulated in HNSCC tissues.ConclusionsThe level of metabolites and the expression of enzymes differ between HNSCC and normal tissues. Reprogramming metabolism in HNSCC provides an energy source as well as structural components, creating a system that offers rapid proliferation, progression, and is less likely to be eliminated.
