Autophagy ensures the lysosome-mediated breakdown and recycling of self-material, as it not only degrades obsolete or damaged intracellular constituents but also provides building blocks for biosynthetic and energy producing reactions. Studies in animal models including Drosophila revealed that autophagy defects lead to the rapid decline of neuromuscular function, neurodegeneration, sensitivity to stress (such as starvation or oxidative damage), and stem cell loss. Of note, recently identified human Atg gene mutations cause similar symptoms including ataxia and mental retardation. Physiologically, autophagic degradation (flux) is known to decrease during aging, and this defect likely contributes to the development of such age-associated diseases. Many manipulations that extend lifespan (including dietary restriction, reduced TOR kinase signaling, exercise or treatment with various anti-aging substances) require autophagy for their beneficial effect on longevity, pointing to the key role of this housekeeping process. Importantly, genetic (e.g., Atg8a overexpression in either neurons or muscle) or pharmacological (e.g., feeding rapamycin or spermidine to animals) promotion of autophagy has been successfully used to extend lifespan in Drosophila, suggesting that this intracellular degradation pathway can rejuvenate cells and organisms. In this review, we highlight key discoveries and recent progress in understanding the relationship of autophagy and aging in Drosophila.

Although death is inevitable, individuals have long sought to alter the course of the ageing process. Indeed, ageing has proved to be modifiable; by intervening in biological systems such as nutrient-sensing, cellular senescence, the systemic environment and the gut microbiome, phenotypes of ageing can be slowed sufficiently to mitigate age-related functional decline. These interventions can also delay the onset of many disabling, chronic diseases, including cancer, cardiovascular disease and neurodegeneration in animal models.Here we examine the most promising chemical interventions to slow ageing, and group them into two tiers based on the robustness of the preclinical, and some clinical, results. We then focus on the potential of the interventions and the feasibility of conducting clinical trials with these chemicals, with the overall aim of maintaining health for longer before the end of life.

Acetylsalicylic acid or aspirin is the most widely used drug globally for its anti-inflammatory characteristics, although little is known about its actions on the central nervous system (CNS). We investigated aspirin's potential neuroprotective effects against paraquat-induced neurotoxicity (PQ) in the present study. Adult male wild-type flies were exposed to a diet containing PQ (3 mM) and/or aspirin (1 μM; 5 μM; 10 μM). Flies fed with PQ reduced locomotion and increased mortality. PQ-induced neurotoxicity has also been associated with a marked decrease in acetylcholinesterase (AChE) activity and lipid peroxidation. Co-exposure to aspirin (5 μM) increased survival, improved motor performance, increased AChE activity, and decreased lipid peroxidation. Our results suggest aspirin's neuroprotective effects, probably due to its lysosomal action and antioxidant characteristics. Thus, we demonstrate that the Drosophila melanogaster model can elucidate basic aspirin mechanisms to assist the evaluations carried out in higher animals to minimize the neurodegenerative effects caused by diseases such as Parkinson's and Alzheimer's.
