Metabolites of the Nitric Oxide (NO) Pathway Are Altered and Indicative of Reduced NO and Arginine Bioavailability in Patients with Cardiometabolic Diseases Complicated with Chronic Wounds of Lower Extremities: Targeted Metabolomics Approach (LC-MS/MS)

Abstract: Objective. The status of metabolites of the nitric oxide (NO) pathway in patients with chronic wounds in the course of cardiometabolic diseases is largely unknown. Yet arginine supplementation and citrulline supplementation as novel therapeutic modalities aimed at increasing NO are tested. Material and Methods. Targeted metabolomics approach (LC-MS/MS) was applied to determine the concentrations of L-arginine, L-citrulline, asymmetric and symmetric dimethylarginines (ADMA and SDMA), and arginine/ADMA and argin… Show more

“…Standard calibration curves were prepared using the following concentration ranges: 5-250 µM for arginine, 0.05-2.5 µM for ADMA and SDMA, 1-50 µM for citrulline, and 0.14-7.0 µM for DMA. The method is characterized by the following intra-and inter-assay CVs: 1.6% and 3.3% (L-arginine), 3.2% and 3.1% (L-citrulline), 1.8% and 5.9% (DMA), 7.5% and 9.4% (ADMA), and 6.4% and 7.1% (SDMA) and was successfully used in patients with hematological malignancies [69], neurodegenerative disorders [70], and cardiometabolic diseases [71].…”

“…As a systematic analyzing tool for profiling metabolites, metabolomics was characterized for high resolution, sensitivity, and throughput in detecting a large population of molecules in various biological samples (8)(9)(10). In the last decade, the advance in metabolomic techniques allowed the identification of numerous individual chemicals in a small volume of biofluid.…”
