Metabolism of indole-3-acetaldoxime in plants

Abstract: Living tissues of diverse plants representing 17 families were infiltrated with indole-3-acetaldoxime (IAAld oxime) in phosphate buffer, pH 6, and incubated for 3 hours at 25°C. Indole compounds were then extracted, separated and identified by paper or thin-layer chromatography (TLC). Indole-3-acetic acid (IAA) was quantitatively determined. Every tissue tested converted the oxime to IAA and tryptophol (T-ol). While accumulation of indole-3-acetonitrile (IAN) was observed in the non-acidic fractions of extract… Show more

“…It should be noted, however, that a direct conversion pathway from IAOx to IAA or indole-3 ethanol has been postulated before (31), and, based on our data, the existence of such a pathway cannot be precluded. Interestingly, this pathway uses IAAld and not IAN as an intermediate from which IAA or indole-3 ethanol is formed by oxidation or reduction.…”

“…IAAld is the precursor of IAA in the Trp-indole-3-pyruvic acid-IAAld-IAA pathway, which may indicate a deregulation of this pathway in the sur2 mutant. However, IAAld has been also postulated to be an intermediate in the Trp-IAOx-IAN-IAA pathway (30,31). Labeling experiments showed that only minute amounts of label were transferred from IAAld to IAA, indicating that no real up-regulation of the Trp-indole-3-pyruvic acid-IAAld-IAA pathway occurred.…”

“…At present, it is not clear how IAOx is converted to IAA. IAN generally has been considered as the primary intermediate between IAOx and IAA, although IAAld has also been proposed (Rajagopal and Larsen 1972). IAN measurements in CYP79B2ox lines and the cyp79B2 and cyp79B3 single and double mutants are consistent with IAN being a direct IAA intermediate.…”

“…Labeling studies have shown that indole-3-acetaldoxime can be converted to IAA with indole-3-acetaldehyde as an intermediate in higher plants (Rajapogal and Larsen, 1972). Accordingly, it has been shown that in the IAA overproducer sur1, the aldehyde oxidase activity is increased (Seo et al, 1998).…”
