Metabolism in tumor microenvironment: Implications for cancer immunotherapy

Abstract: Tumor microenvironment is a special environment for tumor survival, which is characterized by hypoxia, acidity, nutrient deficiency, and immunosuppression. The environment consists of the vasculature, immune cells, extracellular matrix, and proteins or metabolic molecules. A large number of recent studies have shown that not only tumor cells but also the immune cells in the tumor microenvironment have undergone metabolic reprogramming, which is closely related to tumor drug resistance and malignant progression… Show more

“…All these alterations contribute greatly to an immunosuppressive TME, thereby impairing the antitumor immune response and further promoting the tumor progression ( Biswas, 2015 ). Targeting metabolic pathways is therefore an immunotherapeutic approach to inhibit the tumor progression via restoring the TME environment ( Shi et al, 2020 ). In the present study, several signaling pathways related to metabolism were mainly enriched in subtype Sub2, for example, tyrosine metabolism, cytochrome p450 metabolism, and arachidonic acid metabolism.…”

“…One way how tumors can influence Th2 cell-mediated immunity to neoplastic cells is by consuming nutrients and thereby starving Th2 effectors. This mechanism was meanwhile indicated for several immune cells within the TME including macrophages, CD8 + T cells, dendritic cells (DCs), and natural killer cells but not yet for Th2 cells (130,(146)(147)(148)(149). Tumors consume high levels of glucose to perform glycolysis.…”

“…While it is well known that cancer cells undergo metabolic reprogramming, as described above, there is a growing recognition that such metabolic alterations also occur in immune cells, which affects the function of immune cells and contribute to tumor immune escape ( 30 32 ).…”
