Metabolism in Retinopathy of Prematurity

Abstract: Retinopathy of prematurity is defined as retinal abnormalities that occur during development as a consequence of disturbed oxygen conditions and nutrient supply after preterm birth. Both neuronal maturation and retinal vascularization are impaired, leading to the compensatory but uncontrolled retinal neovessel growth. Current therapeutic interventions target the hypoxia-induced neovessels but negatively impact retinal neurons and normal vessels. Emerging evidence suggests that metabolic disturbance is a signif… Show more

“…Both of the mechanisms can promote the release and formation of neoangiogenesis mediators induced by hypoxia. Long term fetal hypoxia and/or ischemia may play a role in the pathogenesis of ROP [1,29] . In addition, premature vaginal delivery can often cause vaginal infection of newborn, which also increases the occurrence of ROP and retinal neovascularization [30][31] .…”

“…Delayed physiologic retinal vascular development causes hypoxia with consequent ischemia that leads to angiogenic signaling with increased expression of VEGF. Hence, vasoproliferation with disordered proliferation into the vitreous occurs (26)(27)(28).…”

“…ROP is related to hypoxia and nutrient deprivation in the maturation of retinal blood vessels, and regulation of retinal metabolism can prevent pathological angiogenesis. Recent research on the metabolic changes of ROP shows that metabolites can serve as biomarkers ( 35 ). Metabolomics affects cell physiology by regulating the genome, epigenome, transcriptome, and proteome ( 36 ).…”
