Metabolism in Invariant Natural Killer T Cells: An Overview

Abstract: Cellular metabolism is critical for generating energy and macromolecules for cell growth and survival. In recent years, the importance of metabolism in mediating T cell differentiation, proliferation, and function has been a hot topic of investigation. However, very little is known about metabolic regulation in invariant natural killer T (iNKT) cells. In this viewpoint, we will discuss what is currently known about immunometabolism in iNKT cells and how these findings relate to CD4 T cells.

“…Invariant natural killer T (iNKT) cells, the main category of natural killer T cells and a subpopulation of mature innate T lymphocytes [ 51 ], were identified as essential players in immunometabolism due to their capacity to respond to self-derived or microbially derived lipid antigens, such us α-galactosylceramide (α-GalCer), which is presented by CD1 molecules on APCs. Similar to these APCs, iNKT cells act as a link between the innate and the adaptive immune system, and they are able to produce the highest amounts of Th1, Th2, Th17, or regulatory cytokines upon activation [ 51 , 52 , 53 , 54 ]. Moreover, there is evidence of communication between cellular metabolic and immune signaling pathways for proper iNKT cell development and function.…”

Thus, functionally, these cells present an anti-inflammatory or prohomeostatic role in disease development or inflammatory activity, with relevant roles in host defense [ 54 ].…”

“…The development of invariant natural killer T (iNKT) cells is more sensitive to changes in mitochondrial electron transport chain function than conventional αβT cells ( 149 ). There have been several informative reviews recently, so we won’t go over those details here ( 150 152 ).…”

“…In particular, the metabolic balance between glycolysis and oxidative phosphorylation (OXPHOS) shapes the differentiation and functions of CD4 + Th cells, Foxp3 + regulatory T cells (Tregs), cytotoxic CD8 + T cells, and macrophages 18 . The development and functions of i NKT cells may also depend on their metabolism: while the evidence to date is quite limited 19 , the thymic development of i NKT cells and their inflammatory cytokine production as mature cells associate with changes in their glycolysis:OXPHOS balance 20 . Moreover, peripheral i NKT-cell functions are regulated by their levels of reactive oxygen species, which are byproducts of aerobic metabolism 21 .…”
