Metabolic transitions define spermatogonial stem cell maturation

Abstract: STUDY QUESTION Do spermatogonia, including spermatogonial stem cells (SSCs), undergo metabolic changes during prepubertal development? SUMMARY ANSWER Here, we show that the metabolic phenotype of prepubertal human spermatogonia is distinct from that of adult spermatogonia and that SSC development is characterized by distinct metabolic transitions from oxidative phosphorylation (OXPHOS) to anaerobic metabolism. … Show more

“…Among all scRNA-seq studies on testicular samples, several studies did original scRNA-seq on underage testicular samples 31 , 55 , 67 , 72 , 79 , 87 . And in some other studies 78 , 88 , previously published underage testicular scRNA-seq data were re-analyzed.…”

“…Metabolites and other substrates can freely diffuse through the whole tissue and developing cords. It seems therefore likely that the concentration of blood‐derived molecules and basal‐specific growth factors are consequently lower, while Sertoli cell‐produced pyruvate and lactate concentrations are higher at the basement membrane relative to the adult testis, where undifferentiated spermatogonia are mainly located after 1 year of development 35 …”

“…Lower panel: Proposed new metabolic development of the SSC maturation (according to results summarizing the metabolism of pre‐pubertal and adult spermatogonia), defined by high aerobic metabolism during development and maintained until maturation of the niche, when an adult‐like metabolic phenotype is established. We refer here to adult SSC only after the adult SSC niche has been developed (image modified from 35 ).…”
