Metabolic Regulation in Pluripotent Stem Cells during Reprogramming and Self-Renewal

Abstract: Small, rapidly dividing pluripotent stem cells (PSCs) have unique energetic and biosynthetic demands compared with typically larger, quiescent differentiated cells. Shifts between glycolysis and oxidative phosphorylation with PSC differentiation or reprogramming to pluripotency are accompanied by changes in cell cycle, biomass, metabolite levels, and redox state. PSC and cancer cell metabolism are overtly similar, with metabolite levels influencing epigenetic/genetic programs. Here, we discuss the emerging rol… Show more

“…Interestingly, hESC-specific downregulated genes were highly enriched in ion/cation transport and metabolism ( Figure 3A). Metabolism was recently proved to be a critical aspect of the unique energetic and biosynthetic programs associated with ESCs' self-renewal [34,35]. Indeed, many actively expressed metabolic genes with reported or unknown roles in ESC maintenance were targeted by SOX2.…”

“…A few recent review articles provide a comprehensive overview of the burgeoning field of metabolism research in stem cells. [2][3][4][5] Even though this field is fairly young, the broad area of "cellular metabolism" refers to a vast and complex interplay of various anabolic and catabolic pathways. Since the recognition that not only do undifferentiated stem cells and mature stem cells have distinct metabolic phenotypes but the metabolic pathways are mechanistically involved in the differentiation processes, research groups have attempted to target a plethora of metabolic pathways in stem cells.…”
