Hibiscus sabdariffa comprises calyx which is rich in flavonoids and numerous phytochemical compounds. Nevertheless, the extraction parameters can influence the recoveries of these bioactive compounds. It is then important to investigate the contributing effects of the process parameters and optimized their conditions to achieve higher recovery yield. Hence, this study was carried out to examine the contributing effects of microwave-assisted extraction (MAE) process factors (irradiation time, ethanol concentration, solvent/sample ratio, temperature, and microwave power) on the yield of total flavonoid (TF) contents from H. sabdariffa calyces using one-variable-at-a-time.The significant MAE process parameters were further optimized using Box Behnken design. At the MAE optimal conditions, the extract was analyzed to identify the flavonoid compounds using liquid chromatography-mass spectrometry quadrupole time of flight. An optimum TF content of 94.32 mg QE/g extract from H. sabdariffa calyces was obtained at the following MAE conditions: Irradiation time of 4 min; ethanol concentration of 52% vol/vol; microwave power of 450 W and solvent/sample ratio 15:1 ml/g. Moreover, 95 flavonoid compounds were tentatively identified in the extract at optimized MAE conditions. Thus, these flavonoids can be further isolated in future studies to unveil embedded potentials of H. sabdariffa calyx extract. Practical applicationThe procedure described the microwave-assisted extraction (MAE) which can serve as a guideline for extracting flavonoids from Hibiscus sabdariffa calyx. Moreover, the liquid chromatography-mass spectrometry quadrupole time of flight analysis provided the developed conditions for identifying flavonoids from the calyces. The results obtained suggested the exact MAE conditions to achieve an optimum yield of flavonoids from H. sabdariffa calyces. Most importantly, the tentatively identified flavonoids can further be isolated and purified in future studies to unveil the embedded biological potentials.

Introduction The herbs Notopterygium incisum (NI) and N. franchetii (NF) are referred to as “Qianghuo” in the Chinese Pharmacopeia and are popular for treatment of certain conditions, including headaches, rheumatoid arthritis and the common cold. Recently, several adulterations of NI and NF have been found in the Chinese herbal market. Objective The aim of this study was to rapidly identify the unique characteristic compounds of NI and NF, to discriminate Qianghuo from its adulterations. Methodology Twenty‐four batches of NI and NF samples with different origins were collected and extracted with methanol. The extracts were analysed using ultrahigh‐performance liquid chromatography‐quadrupole time‐of‐flight tandem mass spectrometry (UHPLC‐QTOF‐MS/MS). Principal component analysis (PCA) and orthogonal partial squared discriminant analysis (OPLS‐DA) were then used to distinguish between NI and NF and to identify their potential characteristic markers. Results Fifty compounds were identified or tentatively characterised according to the retention time, m/z value and MS/MS fragment analysis. Six compounds were selected as potential markers of NI and NF by PCA and OPLS‐DA. They were successfully applied to authenticate 17 kinds of Chinese patent medicines containing Qianghuo. The markers could not be detected in three of the Chinese patent medicines, indicating that they were counterfeit products. Conclusion The UHPLC‐QTOF‐MS/MS coupled with the multivariate analysis method could discriminate NI and NF from their adulterations. Moreover, the data clearly demonstrated significant differences in the chemical compositions of NI and NF. Further research is needed to examine the relationship between therapeutic efficacy and the chemical constituents of NI and NF.

Microwave-assisted extraction (MAE) of phenolic compounds from Chromolaena odorata leaves was investigated using one-factor-at-a-time (OFAT) and two-level factorial design. The MAE parameters studied were irradiation time (A: 1–5 min); microwave power level (B: 400-800 W); extraction temperature (C: 60–80 °C); solvent/feed ratio (D: 8:1–14:1 mL/g); and ethanol concentration (E: 20–60% v/v). The optima yields of TPC and TFC were 56.13 mg GAE/g d.w. and 44.78 mg QE/g d.w., respectively were achieved from MAE of C. odorata leaf at irradiation time of 2 min, microwave power of 600 W, temperature of 60 °C, solvent:feed ratio of 10:1 mL/g, and ethanol concentration of 40% v/v through one-factor-at-time (OFAT) experimental trials. The results obtained from a two-level factorial design experiments reflected that only ethanol concentration (20–60% v/v), irradiation time (1–5 min) and microwave power level (400–800 W) had significant effects on the yields of total phenolic content (TPC) and total flavonoid content (TFC) from C. odorata leaves (p < 0.05). However, temperature and solvent/feed ratio was not significant. In addition, the interactions AB (irradiation time and microwave power) and AE (irradiation time and ethanol concentration) contributed greatly to the recovery yields.
