Aflatoxin B1 is one of the most important mycotoxins due to its hepatotoxic and carcinogenic effects on some fishes. This study was conducted to investigate the defensive role of curcumin and garlic on phase I of the biotransformation system in the liver of aflatoxicosis in Oreochromis niloticus in relation to the therapeutic properties by studying hepatic histopathology and cytochrome P450-CYP1A gene expression using real-time polymerase chain reaction (RT-PCR). Although varieties of methods have been used for gene expression quantification and are still used today, real-time RT-PCR method has achieved prominence in recent years. One hundred and eighty (180) fish were equally divided into 10 groups. T1 was the negative control; fish in T2 were injected intraperitonealy (IP) with AFB1 at a rate of 6 mg kg -1 B.W. and fed on a basal diet. Groups T3-T6 were fed with garlic (T3 & T4) and curcumin (T5 & T6) at 10 & 20 g kg -1 diet, respectively. Groups T7-T10 injected IP with AFB1 and fed on both garlic and curcumin at the different concentrations (10 & 20 g kg -1 diet). AFB1 reduced the HSI values in aflatoxicosis fish. Feeding a low dose of garlic led to significant improvement in the HSI values. Histologically, hepatic lesions were observed in the AFB1 injected groups; damage was reduced when fish were fed on garlic and curcumin diets at a concentration 10 g kg -1. The highest induction of CYP1A expression was observed in the curcumin groups followed by AFB1. The over expression of CYP1A in T2 was alleviated by the garlic. Therefore, it was found that garlic enhances the detoxification of AFB1 through the suppression of CYP1A in the liver.

This study was designed to establish if Curcumin (CM) alleviates Aflatoxin B1 (AFB1)-induced hepatotoxic effects and to determine whether alteration of the expression of cytochrome P450 (CYP450) isozymes is involved in the regulation of these effects in chick liver. One-day-old male broilers (n = 120) were divided into four groups and used in a two by two factorial trial in which the main factors included supplementing AFB1 (< 5 vs. 100 μg/kg) and CM (0 vs. 150 mg/kg) in a corn/soybean-based diet. Administration of AFB1 induced liver injury, significantly decreasing albumin and total protein concentrations and increasing alanine aminotransferase and aspartate aminotransferase activities in serum, and induced hepatic histological lesions at week 2. AFB1 also significantly decreased hepatic glutathione peroxidase, catalase, and glutathione levels, while increasing malondialdehyde, 8-hydroxydeoxyguanosine, and exo-AFB1-8,9-epoxide (AFBO)-DNA concentrations. In addition, the mRNA and/or activity of enzymes responsible for the bioactivation of AFB1 into AFBO—including CYP1A1, CYP1A2, CYP2A6, and CYP3A4—were significantly induced in liver microsomes after 2-week exposure to AFB1. These alterations induced by AFB1 were prevented by CM supplementation. Conclusively, dietary CM protected chicks from AFB1-induced liver injury, potentially through the synergistic actions of increased antioxidant capacities and inhibition of the pivotal CYP450 isozyme-mediated activation of AFB1 to toxic AFBO.

A Low-Cost Label-Free AFB1 Impedimetric Immunosensor Based on Functionalized CD-Trodes

This work describes the investigation of a label-free immunosensor for the detection of aflatoxin B 1 (AFB 1 ). CD-trodes (electrodes obtained from recordable compact disks) were used as low-cost and disposable transducers after modification with a self-assembled monolayer (SAM) of lipoic acid. The anti-aflatoxin B 1 antibody was immobilized via EDC/NHS activation, followed by blocking with bovine serum albumin and immunoassays with AFB 1 . The optimization of analytical parameters and the detection were carried out using electrochemical impedance measurements. Using chemometric tools, the best conditions for the immunosensor development were defined as: anti-AFB 1 antibody at 1:2000 dilution and surface blocking with 0.5% bovine serum albumin, both incubated for 1 h, and antibody-antigen immunoreaction for 30 min. The impedimetric immunosensor showed a linear range from 5 × 10 −9 to 1 × 10 −7 mol·L −1 (1.56-31.2 ng·mL −1 ), limit of detection and limit of quantification, respectively, 3.6 × 10 −10 and 1.1 × 10 −9 mol·L −1 (0.11 and 0.34 ng·mL −1 ). The proposed immunosensor was applied to analyze peanut samples.
