Metabolic and Genetic Properties of Petriella setifera Precultured on Waste

Abstract: Although fungi that belong to Petriella genus are considered to be favorable agents in the process of microbial decomposition or as plant endophytes, they may simultaneously become plant pests. Hence, nutrition factors are supposed to play an important role. Therefore, it was hypothesized that Petriella setifera compost isolates, precultured on three different waste-based media containing oak sawdust, beet pulp (BP) and wheat bran (WB) will subsequently reveal different metabolic properties and shifts in genet… Show more

“…The C-compounds utilization patterns were prepared using heatmap graphs. The enzyme activities, Average Well Color Development (AWCD) [147], Average Well Density Development (AWDD) [148], Richness (the number of utilized carbon sources), number of OTU's (operational taxonomic units) and Shannon index (H) [149] differences between the treatments were assessed with appropriate statistical tests. The analysis of variance (ANOVA) regarding the soil type and applied fertilization and a post-hoc Tukey test were used for significant differences calculation when ANOVA assumptions were met.…”

“…The most consistent readings allowing good resolution among the treatments came from the 120 incubation hour Biolog Plates and these were used in the analyses. Based on absorbance measurements the average well colour development (AWCD) [32] or average well density development (AWDD) [33] were determined. These parameters are factors of the overall metabolic activity of environmental microorganisms.…”

“…An array of substrates on a microtiter plate was used to assess the exoenzymatic capacity of the tested fungi, since it is impossible to accurately mimic the in chemical environment of the plant inside the plant host. The method of PM that emerged has been proven to have a realistic potential of providing a high throughput of information about the phenotypes of microbial isolates [ 21 , 22 , 23 , 24 ].…”
