Metabolic Activities of Akinetes of the Cyanobacterium Anabaena doliolum: Oxygen Exchange, Photosynthetic Pigments and Enzymes of Nitrogen Metabolism

Abstract: All vegetative cells of Anabaena dolwlum differentiated into akinetes. These showed respiratory O2 uptake but lacked photosynthetic pigments and O2 evolution. Activities of nitrogenase, glutamine synthetase, nitrate reductase and aspartate dehydrogenase were totally absent, while glutamate dehydrogenase showed extremely low activity. Glutamate-oxaloacetate transaminase activity was similar to that in whole filaments but alanine dehydrogenase and glutamatepyruvate transaminase activities were much lower. Thus, … Show more

“…3, GPT activity was rather low in the akinetes (5 nmol product formed min"' mg"p rotein) as compared to whole filaments (15 nmol product formed min"' mg"' protein). However, during akinete germination the GPT activity increased sharply, reaching an activity of 22 nmol product formed min"' mg"' protein after 24 h and then slowly declining to an activity similar to that of wbole filaments by 96 h. NR, AsDH, GS (both transferase and biosyntbetic activities) and nitrogenase were all undetectable in the akinetes (Rao et at., 1984). The NR activity appeared 24 h after the initiation of germination and its appearance coincided witb tbat of photosynthesis; it continued to increase over the first 60 h reaching a maximum of 16 nmol product formed min"'mg"' protein but then declined and stabilized at an activity similar to that of whole filaments (6-5 nmol product formed min"' mg"' protein).…”

“…Akinetes of A. dolioum lack the activities of nitrogenase and enzymes of inorganic nitrogen metabolism (Rao et at., 1984). Since the akinetes nevertheless, germinate in nitrogen-free medium we investigated the sequence of appearance of these enzymes.…”

“…The akinetes were yellow and the visual change in the colour of the cultures from blue-green to brown greatly facilitated the recognition and handling of populations of mature akinetes. The akinetes had a respiration rate of 34 9-37-6 nmol O^ uptake mg"' protein min"' in comparison with 85.8-87.2 nmol O.^ uptake mg ' protein min ' in whole heterocystous filaments, indicating a lower rate of respiration as found in akinetes of Nostoc PCC 7524 (Chauvat et al, 1982) and Anabaena doliolum (Rao . "/ al., 1984).The akinetes showed no photosynthetic O^ evolution although whole filaments had a rate of 374 to 377 nmol Oj evolved mg' protein min"'.…”
