MesP1: a novel basic helix-loop-helix protein expressed in the nascent mesodermal cells during mouse gastrulation

Abstract: A subtractive hybridization strategy was used to isolate putative genes involved in the development of mouse primordial germ cells (PGC). Complimentary DNA was amplified on RNA isolated from the base of the allantois where PGC are located in the 7.5 days post coitum (dpc) mouse embryo. It was then subtracted by hybridization with cDNA amplified on RNA of the anterior region where PGC are absent. A novel gene thus isolated is designated as Mesp1 and encodes a possible transcription factor MesP1 containing a bas… Show more

“…4 A-C). One of which is Mesp1, which is essential for early mesodermal differentiation [ 38 ]. Indeed, Mesp1 was not only higher expressed in WT cells, the gene was also more strongly decorated with H3K27me3 in Nox4 −/− cells (orange arrow as indicators).…”

“…Mck-Cre ablated Rnf20 in cardiac and skeletal muscle cells beginning at E13.5 40 (Figure S4). Notably, the conditional deletion of Rnf20 before chamber formation by Mesp1-Cre (activated at embryonic day 6.5) 44 (Figure S5A) was embryonically lethal (Table S2), and the mutant embryos displayed defects in heart morphogenesis as early as E9.5 (Figure S5B). In contrast, the depletion of RNF20 by Mck-Cre ( cKO ) did not affect the Mendelian ratio of each genotype at postnatal day 1 (P1) (Table S1), but it caused the mice to exhibit significantly lower body weights and sudden death within 8 weeks (P56) of birth (Figure 2A and 2B).…”
