Meroterpenoids and Isocoumarinoids from a Myrothecium Fungus Associated with Apocynum venetum

Abstract: Four new meroterpenoids 1–4 and four new isocoumarinoids 5–8, along with five known isocoumarinoids (9–13), were isolated from the fungus Myrothecium sp. OUCMDZ-2784 associated with the salt-resistant medicinal plant, Apocynum venetum (Apocynaceae). Their structures were elucidated by means of spectroscopic analysis, X-ray crystallography, ECD spectra and quantum chemical calculations. Compounds 1–5, 7, 9 and 10 showed weak α-glucosidase inhibition with the IC50 values of 0.50, 0.66, 0.058, 0.20, 0.32, 0.036, … Show more

“…was investigated. Compounds 76, 81, 85, and 87 exhibited inhibitory potential towards the Saccharomyces cerevisiae expressed human-sourced α-glucosidase recombinant with IC 50 s 0.37, 0.32, 0.036, and 0.026 mM, respectively compared to acarbose (IC 50 0.47 mM)[103]. Compounds 165, 167, 174, and 218 biosynthesized by Leptosphaena maculans did not show any inhibitory effects against α-glucosidase…”

“…The inhibitions of the compounds against α-glucosidase from Saccharomyces cerevisiae were assayed by reported method (Xu et al, 2018). The testing compounds were dissolved in dimethyl sulfoxide (DMSO) to obtain stock solution (10 mM) and then diluted into the concentrations by PBS (pH 6.8), while α-glucosidase (2.0 U/mL, Sigma), 4-nitrophenyl-α-Dglucopyranoside (PNPG, 2.5 mM, Macklin), Na 2 CO 3 (0.2 M), and acarbose (2.5 mg/mL, Sigma) were directly dissolved in PBS.…”

“…Studies of the fungus Myrothecium sp. OUCMDZ-2784 associated with the salt-resistant medicinal plant, Apocynum venetum (Apocynaceae) (the estuary of Yellow River, China), afforded four new meroterpenoids, myrothecisins A-D (366-369), which displayed a weak α-glucosidase inhibitory effect (IC 50 = 0.50, 0.66, 0.058, and 0.20 mM vs. 0.47 mM for acarbose) [117].…”
