Mercaptoethanol-Stable Antibody Test Predicting Hemolytic Disease of the Newborn Due to ABO Incompatibility

Abstract: Maternal sera were titrated after 2-mercaptoethanol treatment for anti-A and anti-B agglutinations by the saline and the anti-globulin methods; the results were expressed in titration scores. Each case was graded into mercaptoethanol-stable antibody (MESA) grade 1 : A and B mothers with compatible infants; 2: mothers with unaffected incompatible infants; 3 : mothers with mild hemolytic disease of the infant, and 4: mothers with severe hemolytic disease of the infant. 925 cases were submitted for the ABO incomp… Show more

“…The identification of red cell antibodies as IgM or IgG immunoglobulins often provides useful information in blood banking (Hasekura, 1974;Moor and Steane, 1976). IgM immunoglobulin may be inactivated by treatment with sulphydryl compounds such as 2-mercaptoethanol (2-ME).…”

“…(a) In our study the titre of the IgG antibodies was determined after delivery, while in Hasekura's study [2] this was done in late pregnancy, and Hasekura observed that in some patients post-partum titres may become lower in relation to those found before delivery.…”

“…Numerous techniques for the detection of IgG antibodies anti-A or -B have been proposed but those based on the neutralization of IgM antibodies by 2-mercaptoethanol seem to give the best results [1][2][3][4][5]. The method described by Reesink et al [4] is particularly attractive since it is simple and less time-consuming than other techniques, while it has proved to be as reliable as other methods.…”
