Mentholation affects the cigarette microbiota by selecting for bacteria resistant to harsh environmental conditions and selecting against potential bacterial pathogens

Abstract: BackgroundThere is a paucity of data regarding the microbial constituents of tobacco products and their impacts on public health. Moreover, there has been no comparative characterization performed on the bacterial microbiota associated with the addition of menthol, an additive that has been used by tobacco manufacturers for nearly a century. To address this knowledge gap, we conducted bacterial community profiling on tobacco from user- and custom-mentholated/non-mentholated cigarette pairs, as well as a commer… Show more

“…Specifically, Acidovorax spp. have been identified in two common brands of cigarettes [ 38 ] and have the capacity to metabolize multiple organic pollutants like those found in cigarette smoke [ 39 ]. Therefore, degradation of tobacco smoke compounds, such as polycyclic aromatic hydrocarbons by Acidovorax spp ., may promote survival of transformed cells and subsequently tumor promotion.…”

“…For the 16S rRNA gene analysis, DNA was extracted from each of the four filter quadrants from the 1 and 0.2 μm filters utilizing an enzymatic and mechanical lysis procedure described in detail elsewhere ( Chopyk et al, 2017a , b ).…”

“…The V3-V4 hypervariable region of the 16S rRNA gene was PCR-amplified and sequenced on the Illumina HiSeq (Illumina, San Diego, CA, United States) utilizing a dual-indexing strategy for multiplexed sequencing developed at the Institute for Genome Sciences ( Fadrosh et al, 2014 ), described in detail elsewhere ( Chopyk et al, 2017a , b ).…”

“…Given that Acidovorax spp. , which have been identified in 2 common brands of cigarettes [38], have the capacity to metabolize multiple organic pollutants, like those found in cigarette smoke, it is plausible that smoking creates an environment that allows this bacterium to outcompete other species for resources and thus survival [39]. Further, these bacteria are capable of metabolizing polycyclic aromatic hydrocarbons, which are tobacco smoke carcinogens that can cause DNA adducts, suggesting they may actually contribute to lung carcinogenesis.…”
