Mendelian Randomization Study on the Putative Causal Effects of Omega-3 Fatty Acids on Low Back Pain

Abstract: Previous observational studies have suggested an important role of omega-3 in low back pain. In the present study, we used a two-sample Mendelian randomization (MR) study to identify the putative causal link between omega-3 and low back pain. A broadly used genome-wide association study (GWAS) (n = 8,866 individuals from European ancestry) was used to select plasma omega-3 genetic instrumental variables (IVs). A previously reported GWAS (4,863 cases and 74,589 controls from European ancestry) for low back pain… Show more

“…The IVW was transformed into a weighted regression of the outcome effects of instrumental variables on exposure effects to obtain an overall estimate of the impact of BMI, WHRadjBMI, WHR and WC on glaucoma risk. Meanwhile, the fixed effect model or random effect model were selected according to the heterogeneity (31,32). The MR-Egger method can still provide unbiased estimators even if pleiotropy exists for all selected instrumental variables.…”

“…Using genetic variants independent of many factors that bias observational studies, Mendelian randomization (MR) studies have many advantages in assessing the causal association between an exposure and an outcome. 8 , 9 , 10 , 11 , 12 , 13 Thus, we used a two‐sample MR study to identify the causal genetic link between COVID‐19 and childhood intelligence.…”

“…On the other hand, large randomized trials, which can provide the best evidence for causation, are expensive and time-consuming. Mendelian randomization (MR) provides an ingenious method for inferring the causal nature of the exposure-outcome relationship by using instrumental variables (IVs), such as single nucleotide polymorphisms (SNPs) ( Gao et al, 2021 ; Zhou et al, 2022 ). Due to the random segregation of alleles at conception, genetic variants are fixed and not affected by environmental risk factors, which minimizes residual confounding and reverse causality.…”
