Membrane Lymphotoxin Is Required for the Development of Different Subpopulations of NK T Cells

Abstract: The development of lymphoid organs requires membrane-bound lymphotoxin (LT), a heterotrimer containing LTα and LTβ, but the effects of LT on T cell function have not been characterized extensively. Upon TCR cross-linking in vitro, splenocytes from both LTα−/− and LTβ−/− mice failed to produce IL-4 and IL-10 due to a reduction in NK T cells. Concordantly, LTα−/− and LTβ−/− mice did not respond to the lipoglycan α-galactosylceramide, which is presented by mouse CD1 to Vα14+ NK T cells. Interestingly, both popula… Show more

“…These mice have significantly reduced levels of Aire [20] and NKT cells [21,22], but relatively normal conventional T-cell numbers. A similar correlation between Aire expression and NKT cell numbers was reported in lymphotoxin-deficient mice [23,24], although the link between the expression of lymphotoxin and Aire-dependent genes has been disputed in more recent studies [25][26][27]. Human patients (and a mouse model) of Omenn syndrome [28,29] also have reduced levels of Aire and NKT cells and it is interesting to note that the severe NKT cell deficit evident in NOD mice coincides with significant morphological abnormalities among Aire 1 medullary cells in the thymus [20].…”

“…Consistent with these findings, cytokine production levels induced by in vivo immunization with the glycolipid ␣-GalCer were found to be reduced in LT␣ 0/0 , LT␤ 0/0 , and LT␤R 0/0 mice but not in LIGHT 0/0 mice (ref. 24 and unpublished data). Nevertheless, the ability of the residual V␣14i NKT cells to produce IFN-␥ (Fig.…”
