Membrane Fusion and the Lamellar-to-Inverted-Hexagonal Phase Transition in Cardiolipin Vesicle Systems Induced by Divalent Cations

Abstract: The polymorphic phase behavior of bovine heart cardiolipin (CL) in the presence of different divalent cations and the kinetics of CL vesicle fusion induced by these cations have been investigated. (31)P-NMR measurements of equilibrium cation-CL complexes showed the lamellar-to-hexagonal (L(alpha)-H(II)) transition temperature (T(H)) to be 20-25 degrees C for the Sr(2+) and Ba(2+) complexes, whereas in the presence of Ca(2+) or Mg(2+) the T(H) was below 0 degrees C. In the presence of Sr(2+) or Ba(2+), CL large… Show more

“…Cardiolipin stimulates Ypt7p-independent fusion of standard-method proteoliposomes (Fig. S5E), consistent with the finding that Mg 2ϩ -cardiolipin can mediate liposome fusion without leakage (70). Does cardiolipin play a role in vacuole fusion in vivo?…”

“…Zinc ions, for example, are known to bind to both charged and uncharged lipids, changing the bilayer effective surface charge density and elastic bending properties with concomitant lipid headgroup dehydration (see Fig. 5B) (25,38). In contrast, whereas Zn 2ϩ is also known to interact with DNA, this interaction alone is insufficient to cause DNA collapse in solution (34).…”

“…Specifically, mitochondrial size is more variable, giant mitochondria more abundant, and, similar to our observations (Figure 8, C-F), cristae morphology is altered in the absence of Taz1p activity. CL is a so-called structural phospholipid capable of adopting lamellal or inverted hexagonal structures depending on the presence of divalent cations (Ortiz et al, 1999). Taz1p, as a transacylase, can actually perform this function in either direction, i.e., it can transfer an acyl chain from PC to MLCL and from CL to lyso-PC (Xu et al, 2006b).…”
