Membrane-bound cytochrome c is an alternative electron donor for cytochrome aa3 in Nitrobacter winogradskyi

Abstract: We purified membrane-bound cytochrome c-550 [cytochrome c-550(m)] to an electrophoretically homogeneous state from Nitrobacter winogradskyi. The cytochrome showed peaks at 409 and 525 nm in the oxidized form and peaks at 416, 521, and 550 nm in the reduced form. The molecular weight of the cytochrome was estimated to be 18,400 on the basis of protein and heme c contents and 18,600 by gel filtration. The N-terminal amino acid sequence of cytochrome c-550(m) was determined to be A-P-T-S-A-A-D-A-E-S-F-N-K-A-L-A-S… Show more

“…Similar membraneassociated C-type cytochromes have recently been described in some bacteria such as CycMin Bradyrhizobium japonicum (Bott et al, 1991), thecssl in Chlorobiumvibrioforme(Okke1s et al, 1992, and the c5500f Bacillussubtilis (von Wachenfeldt & Hederstedt, 1993). A newly reported membrane-associated C-type cytochrome in Nitrobacter winogradskyi may also be similar to cyt cy based on its proposed function and membrane location (Nomoto et al, 1993). Thus, membrane-associated electron carriers such as cyt cy may be more widely spread and may play a larger role in electron transport chains than has previously been suspected.…”

“…DNA hybridization to coxA, coxB, and coxC probes revealed multiple copies of the cytochrome oxidase operon (11). Both soluble and membrane-bound class IA/B cytochrome c-550 proteins were purified and shown to be electron donors for Cox (67,87,102). Among the c-type cytochrome genes identified, orthologs for both cytochrome c-550 proteins (c-550s, nwi2582, and c-550m, nwi0712) were located.…”

“…They are known for their heterotrophy (Bock 1976), mixotrophy (Smith & Hoare 1968), heterogeneity among species (Navarro et al 1992), anaerobic growth by dissimilatory nitrate reduction (Freitag et al 1987, Bock et al 1988. Freitag & Bock 1990, as well as alternate respiratory chains (Bock et al 1986, Nomoto et al 1993. The inherent metabolic diversity of nitrite oxidizers may be the key to their phototolerance.…”
