Membrane Association of Glutathione S-Transferase mGSTA4-4, an Enzyme That Metabolizes Lipid Peroxidation Products

Abstract: Lipid peroxidation products have signaling functions and at higher concentrations are toxic and may trigger cell death. The compounds are metabolized predominantly by glutathione S-transferases exemplified by mGSTA4-4, an enzyme highly efficient in glutathione conjugation of 4-hydroxyalkenals, and possessing glutathione peroxidase activity toward phospholipid hydroperoxides. mGSTA4-4 belongs to the predominant group of "canonical" glutathione S-transferases that are soluble and generally localized in the cytop… Show more

“…In the wild type form, SOD dismutates superoxide to oxygen and water, hence reducing the levels of oxidative stress and protecting proteins, lipid and DNA from the toxic superoxide molecule [48]. In the present study a significant increase in the expression of SOD1 and significant increase in SOD enzymatic activity in the brain from canines that had undergone a combination of both treatment with antioxidant diet and a program of behavioral enrichment compared to age matched controls were found Glutathione-S-transferase (GST) catalyzes the conjugation of a number of exogenous and endogenous compounds such as 4-hydroxynonenal (HNE) or malondialdehyde (MDA) with glutathione inactivating the toxic products of oxygen metabolism [98]. Hence, GST plays a critical role in cellular protection against oxidative stress.…”

“…Membrane association of GSTs could be important for conjugation of hydrophobic toxins partitioned in the membrane (54). It is interesting that murine GSTA4-4, an enzyme highly efficient in the GSH conjugation of 4-hydoxyalkenals, is also closely associated with the plasma membrane (55). In addition, GSTs from the pi and the mu classes have recently been shown to be associated with goat sperm plasma membrane, where in addition to a role in detoxification, these proteins function as gamete recognition molecules (56).…”

“…Erythrocytes are excellent models for studying of the interactions of xenobiotics with biomembranes, 23) due to alteration in membrane permeability, hence, the use of both plasma and erythrocytes in this study. Also, polyunsaturated fatty acids in erythrocyte membranes, the presence of iron in the heme and oxygen enhance the production of oxidative stress.…”
