Melt Extrusion in Drug Delivery: Three Decades of Progress

Abstract: This chapter appraises the role of melt extrusion as a solubilization and bioavailability enhancement technique. The introductory chapter highlights major aspects of hot melt extrusion (HME) technology as applied in the pharmaceutical industry, particularly processing techniques, material considerations, recent innovative applications of melt extrusion in drug delivery system design, and a review of current HME-based formulations (marketed or under commercial development). The chapter also focuses on key devel… Show more

“…This difference in the operation becomes particularly important when processing thermolabile drugs (20). Compared to the SSE, the intermeshing co-rotating of the TSE provides better mixing, producing a more homogeneous solid containing finely distributed and dispersed active compounds, higher kneading potential, and better melt temperature control (5). Therefore, there is a lower overheating In an extrusion process, the dimensions of the screws, which are given in terms of the length-to-diameter ratio (L/ D), positioned inside the stationary cylindrical barrel is an important characteristic to consider regardless of whether the extrusion equipment is an SSE or TSE.…”

“…Some notable examples include Kaletra®, Onmel®, Incivek®, Zelboraf®, Kalydeco®, and Noxafil®, among others (1,15). There are a number of reasons why ASDs are becoming the preferred technology to enable the delivery of poorly watersoluble drugs.…”

“…Clearly, HME has been identified as an important and significant process to further enhance drug solubility and solid-dispersion production. [5][6] Hot-melt extrusion (HME) processing were come in action in the early 1930s, and during that time, and became the most widely applied processing technology in the plastic, rubber, and food industries. The application of HME expanded to the pharmaceutical industry at the beginning of the seventies for formulation and product development as well as manufacturing.…”
