Melatonin suppresses aromatase expression and activity in breast cancer associated fibroblasts

Abstract: The main biological active substance secreted by the pineal gland, melatonin (MLT), counteracts the effects of estrogens in breast cancer via exerting a number of its own oncostatic properties. Recent studies of postmenopausal women have identified that the major metabolite of MLT is statistically significantly associated with a lower risk of developing breast cancer. While MLT production decreases with age, breast cancer risk, however, increases with age and obesity. We hypothesize that MLT inhibits estrogen … Show more

“…One of the other abilities of melatonin is the inhibition of aromatase expression and activity in both invivo and invitro [44,[49][50][51][53][54][55]. With respect to this investigation, melatonin not only interacts with the estrogen-responsive pathway, but also inhibits local synthesis of estrogen through preventing the conversion of androgens to estrogen [44,[50][51][52][53][54].…”

“…Aromatase is one of the main enzymes in the conversion of androgens to estrogen [49][50][51]. In light of the role of estrogen in breast cancer, it is obvious that aromatase can play a crucial role in breast cancer progression [49][50][51].…”

“…Eventually lower levels of melatonin in aging women may increase the risk of progressing ER-alpha-positive breast cancer through a decreased ability to suppress CYP19A1 expression. Subsequent excess local estrogen is derived from breast adipose fi broblasts which are the main local source of estrogen in breast tumors of postmenopausal women (Knower et al 2012 ). At physiologic concentrations, melatonin modulates the synthesis and transformation of biologically active estrogens in human breast cancer cells, through the inhibition of enzymes involved in the estradiol formation in breast cancer cells, sulfatase and 17beta-hydroxysteroid dehydrogenase type 1 (Gonzalez et al 2008 ).…”
