Melatonin sees the light: blocking GABA‐ergic transmission in the paraventricular nucleus induces daytime secretion of melatonin

Abstract: Despite a pronounced inhibitory effect of light on pineal melatonin synthesis, usually the daily melatonin rhythm is not a passive response to the surrounding world. In mammals, and almost every other vertebrate species studied so far, the melatonin rhythm is coupled to an endogenous pacemaker, i.e. a circadian clock. In mammals the principal circadian pacemaker is located in the suprachiasmatic nuclei (SCN), a bilateral cluster of neurons in the anterior hypothalamus. In the present paper we show in the rat t… Show more

“…3). The SCN releases GABA to inhibit the sympathetic input to the pineal gland during the daytime while this inhibition is released during the night (Kalsbeek et al 2000). In addition, the SCN sends a constant glutamatergic stimulatory input to the pineal gland which is overwhelmed by the inhibitory mechanism during the night (Perreau- Lenz et al 2004).…”

“…Properties of the circadian pacemaker have also been studied by monitoring in vivo secretion of circadian hormones such as melatonin using a microdialysis probe inserted into the pineal gland (Azekawa et al, 1990(Azekawa et al, , 1991Drifhgout et al, 1993;Barassin et al, 1999;Kalsbeek et al, 2000), the site of melatonin production (Borjigin et al, 1999). Melatonin production in the pineal gland is controlled by the circadian pacemaker in the SCN via a multi-synaptic neuronal pathway ( Figure 1A; Kennaway, 1997).…”

“…In earlier experiments, microdialysis sampling and analyses of melatonin were carried out in vivo for 1-4 days in individual animals, and the results were averaged for a group of animals (Azekawa et al, 1990(Azekawa et al, , 1991Drifhgout et al, 1993;Kalsbeek et al, 2000). Inter-individual variations of the circadian pacemaker activity were difficult to define.…”

“…Furthermore, light inhibits melatonin secretion through the ANS (142) and light has been reported to affect glucocorticoid secretion, although some studies describe increased glucocorticoid levels due to bright light (143,144) , whereas another study describes decreased glucocorticoid levels (145,146) . These inconsistent findings might be related to the duration, intensity or timing of the light exposure.…”
