Soil salinity severely inhibits leaf photosynthesis and limits agricultural production. Red to far-red light ratio (R/FR) affects leaf photosynthesis under salt stress, however, its regulation mechanism is still largely unknown. This study investigated the effects of different R/FR on plant growth, gas exchange parameters, photosynthetic electron transport, Calvin cycle and key gene expression under salt stress. Cucumber seedlings were exposed to four treatments including 0 mM NaCl and R/FR=7 (L7, control), 0 mM NaCl and R/FR=0.7 (L0.7), 80 mM NaCl and R/FR=7 (H7) and 80 mM NaCl and R/FR=0.7 (H0.7) for 9 days in an artificial climate chamber. The results showed that compared to L7 treatment, H7 treatment significantly reduced relative growth rate (RGR), CO2 assimilation rate (Pn), maximum photochemical efficiency PSII (Fv/Fm), most JIP-test parameters and total Rubisco activity, indicating that salt stress severely inhibited photosynthetic electron transport from PSII to PSI and blocked Calvin cycle in cucumber leaves. However, these suppressions were effectively alleviated by low R/FR addition (H0.7 treatment). Compared to H7 treatment, H0.7 treatment significantly increased RGR and Pn by 209.09% and 7.59%, respectively, enhanced Fv/Fm, maximum quantum yield for primary photochemistry (φPo), quantum yield for electron transport (φEo) and total Rubisco activity by 192.31%, 17.6%, 36.84% and 37.08%, respectively, and largely up-regulated expressions of most key genes involved in electron transport and Calvin cycle. In conclusion, low R/FR effectively alleviated the negative effects of salt stress on leaf photosynthesis by accelerating photosynthetic electron transport from PSII to PQ pool and promoting Calvin cycle in cucumber plants. It provides a novel environmentally friendly light-quality regulation technology for high efficiency salt-resistant vegetable production.

Background Soil salinization severely affects plant photosynthesis and reduces crop yield and quality. Therefore, we investigated the structure and function of photosystem I (PSI) and photosystem II (PSII) of tomato seedlings [Zhongshu No.4 (salt-sensitive) and Jinpeng No.3 (salt-tolerant)] after salt stress treatment were analyzed using rapid chlorophyll fluorescence kinetics and 820-nm transmission kinetics. Moreover, the activity and transcript level of enzymes related to Calvin cycle and sucrose metabolism were investigated. Results The structure and function of PSI and PSII were suppressed in both tomato varieties under salt stress as evidenced by Electron transfer chains are inhibited from transferring electrons, photosynthetic reaction centers are damaged, and energy flow distribution is disrupted. In addition, salt stress significantly inhibited the carbon assimilation efficiency of both tomato varieties as indicated by decrease in the activities of Rubisco (initial and total), RCA, PGK, FBPase, GAPDH, and FBA and transcript level and promoted sugar accumulation. Compared with salt-sensitive Zhongshu No.4, the photosynthetic apparatus and carbon metabolism of salt-resistant Jingpeng No.3 were much more tolerant to salt treatment. Conclusion Jingpeng No.3 had a higher electron transfer efficiency. The donor side and acceptor side of PSII, the integrity of the thylakoid, and the oxidized and redox state of PSI were less inhibited by salt stress. Meanwhile, the activation of photosynthetic protection mechanism increased the utilization of energy for photochemical reactions, decreased the excitation pressure of RC and led to a smoother energy flow. Improved carbon assimilation efficiency and sucrose metabolism efficiency. Therefore, Jinpeng No.3 has salt tolerance.

Soil salinity is a severe threat to agricultural production. Most saline soils turn alkaline, increasing the soil pH and, in turn, hampering the growth and development of crops. In this study, the effects of a foliar spray of melatonin (MT; 100 μmol·L−1) on the pH of the root environment, growth of tomato seedlings, endogenous MT levels, rapid chlorophyll fluorescence induction kinetics, and key enzymes of the Calvin cycle under alkaline (60 mmol·L−1 NaHCO3) stress were studied in Riegel 87-5 tomatoes. The results revealed that the growth and photosynthesis of tomato seedlings were inhibited by increased pH in the root environment under alkali stress; however, the application of exogenous MT reduced the pH of the root environment, alleviated the inhibition of growth of tomato seedlings under alkali stress, increased the content of photosynthetic pigments, alleviated the damage of the donor and acceptor sides of the photosynthetic electron transport chain, increased the activity and efficiency of photosynthetic electron transport, and optimized the share of the light energy allocated to PSII reaction centers. Increased expression levels of Calvin-cycle enzymes, including fructose-1,6-bisphosphate aldolase (FBA), fructose-1,6-bisphosphate esterase (FBP), and phosphoglycerate kinase (PGK), led to enhanced photosynthetic performance in tomato seedlings. Exogenous MT boosted endogenous MT levels and stimulated the production and secretion of organic acids in the root system. This regulation of organic acid content reduced the environmental pH in the inter-root zone, alleviating the damage caused by alkali stress. This study indicated that the exogenous administration of MT may mediate an increase in endogenous MT levels, regulate the efficiency of photosynthesis and root pH levels, and play a crucial role in mitigating injury caused by alkali stress in tomato seedlings.
