Melatonin Prevents Brain Damage and Neurocognitive Impairment Induced by Plasmodium Berghei ANKA Infection in Murine Model of Cerebral Malaria

Abstract: Cerebral malaria is characterized by permanent cognitive impairments in Plasmodium-infected children. Antimalarial therapies show little effectiveness to avoid neurological deficits and brain tissue alterations elicited by severe malaria. Melatonin is a well-recognized endogenous hormone involved in the control of brain functions and maintenance of blood-brain barrier integrity. The current study has evaluated the effect of melatonin on the histological alterations, blood-brain barrier leakage, and neurocognit… Show more

“…Body weight variation and survival rates curve were evaluated throughout the experimental period. The evolution of the infection was also monitored daily by determining the parasitemia by Giemsa-stained slides obtained from mice tail blood as described by Ataide et al [ 25 ]. Animals positive to CM show symptoms as poor reflexes, roll-over, ataxia, limb paralysis, seizures and coma between 6- and 10-days post infection.…”

“…Mice were inoculated intraperitoneally (i.p.) with 10 6 parasitized red blood cells (pRBCs), suspended in 200 μ l of phosphate-buffered saline (PBS; pH 7.4) as previously described by Ataide et al [ 23 ]. Treatments with GSH (8 mg/kg/day diluted in 100 μ l PBS) or vitamin C (8 mg/kg/day diluted in 100 μ l PBS) were performed by i.p.…”

“…The brains were then removed and placed in a Petri dish to assess the presence of vascular extravasation in the animal's encephalic microvasculature, performed with the naked eye and under a magnifying glass to capture images. Quantification of extravasated dye was performed as described by Ataíde et al [30].…”
